DNA damage response at telomeres boosts the transcription of SARS-CoV-2 receptor ACE2 during aging

Sara Sepe et al.

Summary: As individuals age, the expression of ACE2 in the lungs increases, making elderly individuals more susceptible to SARS-CoV-2. Telomere damage triggers DDR activation, leading to an upregulation of ACE2 and potentially increasing susceptibility to infection in the elderly.

ALT-FISH quantifies alternative lengthening of telomeres activity by imaging of single-stranded repeats

Lukas Frank et al.

Summary: This study introduces a new method called ALT-FISH to quantitate ALT activity in single cells by detecting the accumulation of single-stranded telomeric DNA and RNA. The method successfully identified ALT in cancer cell lines and primary tissue sections from different tumor entities, and it is suitable for high-throughput applications, which will facilitate screening for ALT-specific drugs.

Telomeric replication stress: the beginning and the end for alternative lengthening of telomeres cancers

Robert Lu et al.

Summary: Telomeres are protein structures that cover the ends of chromosomes. The DNA at telomeres contains repetitive sequences that are difficult to replicate. Structural abnormalities at ALT telomeres increase replication stress, leading to stalling and collapse of replication forks. This DNA damage stimulates recombination-mediated repair and break-induced telomere synthesis. The coordination between replication stress and DNA repair regulates telomere length in ALT cells, and can be manipulated experimentally. Inducing replication stress may selectively disrupt the viability of ALT cancer cells.

RAD51AP1 regulates ALT-HDR through chromatin-directed homeostasis of TERRA

Nicole Kaminski et al.

Summary: In this study, the researchers found that RAD51AP1 interacts with TERRA and utilizes it to generate D and R-loop HR intermediates. RAD51AP1 binds to and stabilizes TERRA-containing R-loops, playing a role in the suppression of TERRA and prevention of TRCs during ALT-HDR. These findings provide insights into the important role of RAD51AP1 in the ALT mechanism and regulation of TERRA.

TERRA and RAD51AP1 promote alternative lengthening of telomeres through an R- to D-loop switch

Tribhuwan Yadav et al.

Summary: This study reveals that the telomere non-coding RNA TERRA can form dynamic telomeric R-loops and contribute to ALT activity. This process is mediated by the formation of G-quadruplexes (G4s), independent of RAD52. The findings shed light on the mechanism of the R-loop to D-loop switch in the ALT pathway and provide new insights into the function and homeostasis of telomeres.

G4 Matters-The Influence of G-Quadruplex Structural Elements on the Antiproliferative Properties of G-Rich Oligonucleotides

Carolina Roxo et al.

Summary: This study demonstrated the potential of G-quadruplexes as anticancer agents. G4s with three G-tetrads and longer loops are more effective in inhibiting cancer cell growth, while highly structured G4s with an extended core containing four G-tetrads show improved cellular uptake and biostability. Understanding the structural features of G4s can lead to the development of novel and more potent G4-based therapeutics with unprecedented anticancer properties.

Summary: The study reveals higher levels of DNA/RNA hybrids and G-quadruplex structures in ALT+ cancer cells, associated with telomere length and DNA damage markers. Nuclear G-quadruplex signals are significantly different in ALT+ tumors compared to TERT+ tumors, serving as a potential biomarker for identifying ALT+ tumors and therapeutic targets in the future.
