CircZNF652 promotes the goblet cell metaplasia by targeting the miR-452-5p/JAK2 signaling pathway in allergic airway epithelia

Xiangzhi Wang et al.

Summary: This study revealed the high expression of circZNF652 in bronchoalveolar lavage fluid of allergic asthma patients and its role in promoting pathological goblet cell metaplasia through activation of the miR-452-5p/JAK2/STAT6 signaling pathway. These findings provide new strategies for the therapeutic intervention of epithelial remodeling in allergic airway inflammation.

CBX4 Regulates Long-Form Thymic Stromal Lymphopoietin-mediated Airway Inflammation through SUMOylation in House Dust Mite-induced Asthma

Shixiu Liang et al.

Summary: This study reveals that small ubiquitin-like modifier (SUMOylation) is enhanced in the airway epithelium of house dust mite-induced allergic asthma. Inhibition of SUMOylation alleviates airway inflammation and lfTSLP expression. Mechanistically, SUMOylation enhances the transcription of MEX-3B through the transcription factor TFII-I, and MEX-3B promotes lfTSLP translation by binding to its mRNA. This finding highlights CBX4 as a potential therapeutic target for lfTSLP-mediated asthma.

Blockade of CBX4-mediated β-catenin SUMOylation attenuates airway epithelial barrier dysfunction in asthma

Shixiu Liang et al.

Summary: This study found that inhibiting SUMOylation can attenuate house dust mite-induced epithelial barrier dysfunction in asthma. The researchers identified CBX4 as the mediator of beta-catenin SUMOylation and showed that silencing CBX4 promotes beta-catenin membrane localization and improves epithelial barrier function.

Interleukin-6 neutralizing antibody attenuates the hypersecretion of airway mucus via inducing the nuclear translocation of Nrf2 in chronic obstructive pulmonary disease

Yuan-Yuan Wei et al.

Summary: Airway mucus hypersecretion is a vital pathophysiologic feature in chronic obstructive pulmonary disease (COPD) patients. The study investigated the mechanisms underlying the action of interleukin-6 neutralizing antibody (IL-6 Ab) in attenuating airway mucus hypersecretion in COPD. It was found that IL-6 Ab can attenuate mucus hypersecretion by inducing nuclear translocation of Nrf2.

PIAS1 and TIF1γ collaborate to promote SnoN SUMOylation and suppression of epithelial-mesenchymal transition

Ayan Chanda et al.

Summary: Studies show that SUMO E3 ligases PIAS1 and TIF1γ collaborate to promote the SUMOylation of SnoN, suppressing EMT in breast cell-derived tissue organoids. These E3 ligases act in an interdependent manner in the regulation process.

PIAS1 modulates striatal transcription, DNA damage repair, and SUMOylation with relevance to Huntington's disease

Eva L. Morozko et al.

Summary: This study reveals the involvement of PIAS1 in DNA damage repair pathways and its connection with transcriptional modulation in neurodegenerative diseases. Knockdown of PIAS1 in HD mice normalizes transcriptional dysregulation and DNA damage repair mechanisms, impacting genomic integrity in neurons. The findings highlight the role of SUMO modifying machinery in DNA damage repair responses and transcriptional modulation in the context of neurodegenerative disease.

Benzo(a)pyrene Enhanced Dermatophagoides Group 1 (Der f 1)-Induced TGFβ1 Signaling Activation Through the Aryl Hydrocarbon Receptor-RhoA Axis in Asthma

Eryi Wang et al.

Summary: The study demonstrates that co-exposure to benzo(a)pyrene (BaP) and dermatophagoides group 1 allergen (Der f 1) enhances airway inflammation in asthma by activating the AhR-RhoA axis, which regulates the expression and signaling activation of TGF beta 1. This suggests a potential therapeutic target for allergic asthma.
