CD83 acts as immediate early response gene in activated macrophages and exhibits specific intracellular trafficking properties

Pia Langguth et al.

Summary: Macrophages are highly plastic cells and understanding their activation and function is key to understanding immunological balance. The CD83 molecule, expressed on activated immune cells and tissue resident macrophages, has been investigated for its potential role in modulating macrophage biology. This study reveals the kinetics and trafficking of CD83 in LPS-activated macrophages, suggesting its importance in early macrophage activation and subsequent cellular fate.

CD83 expressed by macrophages is an important immune checkpoint molecule for the resolution of inflammation

Katrin Peckert-Maier et al.

Summary: This study identifies CD83 as a marker for IL-4 stimulated pro-resolving M phi and shows that CD83 is important for the phenotype and function of these M phi. CD83-deficient M phi exhibit reduced phagocytic capacity, altered STAT-6 phosphorylation pattern, increased production of pro-inflammatory mediators, enhanced stimulation of T cell proliferation, and altered tissue reconstitution after wound infliction.

Chemokine-Driven Migration of Pro-Inflammatory CD4+ T Cells in CNS Autoimmune Disease

Aaron H. S. Heng et al.

Summary: Pro-inflammatory CD4(+) T helper cells play a crucial role in autoimmune diseases. Recent research has revealed the heterogeneity of chemokine receptors expressed by these cells, which are responsible for recruiting different subsets of pro-inflammatory Th cells in autoimmune inflammation. Understanding these receptors may lead to potential therapeutic targets for intervention.

Dissection of artifactual and confounding glial signatures by single-cell sequencing of mouse and human brain

Samuel E. Marsh et al.

Summary: Enzymatic dissociation during single-cell sequencing experiments can lead to aberrant gene expression, especially in microglia. A validated protocol is presented to preserve in vivo transcriptional profiles and cell-type diversity and yield. Similar gene expression changes are observed in postmortem human brain tissue, and these changes can be induced by exposure to elevated temperatures ex vivo.

Cognate microglia-T cell interactions shape the functional regulatory T cell pool in experimental autoimmune encephalomyelitis pathology

Zhana Haimon et al.

Summary: Microglia, the brain macrophages, play a critical role in brain development and homeostasis. They interact with T cells through antigen presentation, costimulation, and coinhibition. In autoimmune neuroinflammation, microglia have significant non-redundant cognate and cytokine-mediated interactions with regulatory T cells.

Summary: This study analyzes 59 scRNA-seq datasets to evaluate the performance of different error models in modeling variation in scRNA-seq data. The results show clear evidence of overdispersion for genes with sufficient sequencing depth, necessitating the use of a negative binomial model. The degree of overdispersion varies widely across datasets, systems, and gene abundances.

Lipoprotein Lipase Regulates Microglial Lipid Droplet Accumulation

Bailey A. Loving et al.

Summary: As individuals age, dysfunctional microglia may contribute to the onset of neurodegenerative diseases through impaired phagocytosis, cholesterol efflux, and enhanced secretion of pro-inflammatory cytokines. Specifically, microglia lacking lipoprotein lipase exhibit a pro-inflammatory lipid profile, increased cholesterol ester content, and reduced cholesterol efflux, leading to excessive accumulation of lipid droplets. Activation of PPAR pathways by agonists can rescue this phenotype, suggesting a potential therapeutic strategy for aging and neurodegenerative diseases.

Quantifying the effect of experimental perturbations at single-cell resolution

Daniel B. Burkhardt et al.

Summary: This study compares treatment and control single-cell RNA sequencing samples more accurately at the single-cell level. By developing a continuous measure of perturbation effects and using graph signal processing to estimate the relative likelihood of observing each cell in different experimental conditions, the study identifies cell populations affected by perturbations and improves the accuracy of identifying clusters of cells enriched or depleted in each condition. The algorithm developed in this study outperforms alternative algorithms in accurately deriving gene signatures from affected cell clusters.

Single-cell RNA-seq analysis reveals compartment-specific heterogeneity and plasticity of microglia

Junying Zheng et al.

Summary: The study revealed overlapping but distinct microglial populations in the cortex and the spinal cord, with varying responses to life span progression and HIV-1 pathogenic protein gp120. This suggests that microglia in different CNS compartments are adapted to fulfill region-specific biological functions in their local environments.

Single-Cell Transcriptomics and In Situ Morphological Analyses Reveal Microglia Heterogeneity Across the Nigrostriatal Pathway

Oihane Uriarte Huarte et al.

Summary: Microglia are immune cells in the CNS that rapidly respond to pathological stimuli to maintain homeostasis, but their reactions may also exacerbate neurological disorders. Studying microglia in healthy tissue can help identify specific subsets that either support or harm the neuronal network. In PD, investigating the heterogeneity of microglial cells in susceptible brain regions like the nigrostriatal pathway is crucial for understanding the disease's progression.

Regulation of microglia population dynamics throughout development, health, and disease

Brady P. Hammond et al.

Summary: The dynamic changes in microglia density, volume, and proliferation in the central nervous system play a crucial role in their function, with fluctuations affecting their ability to respond effectively to various stimuli. Microglia exhibit a robust proliferative capacity during development and in neurodegenerative diseases, but this capacity is largely absent at homeostasis. Proper microglial function in different states is heavily dependent on density fluctuations primarily induced by proliferation.

Characterization of microglial transcriptomes in the brain and spinal cord of mice in early and late experimental autoimmune encephalomyelitis using a RiboTag strategy

Shaona Acharjee et al.

Summary: The study revealed regional heterogeneity in microglial transcriptomes in the brain and spinal cord of animals with EAE, with the most profound gene expression changes observed in the spinal cord of symptomatic animals. Canonical pathway analysis showed alterations in neuroinflammatory pathways, immune functions, and cell division in both pre-onset and symptomatic stages of EAE. The research also highlighted a continuum of pathways from pre-onset to symptomatic stages, indicating temporal heterogeneity in microglial gene expression patterns that may aid in understanding the mechanisms underlying various symptoms of MS.

A Unique Type of Highly-Activated Microglia Evoking Brain Inflammation via Mif/Cd74 Signaling Axis in Aged Mice

Chenghao Jin et al.

Summary: The study investigated the senescence-associated alterations of microglia in the aged brain at the single cell level, revealing six subgroups of microglia with unique functions. Among them, a unique type of highly activated microglia (HAM) was observed only in aged mice. Mechanistically, neuron-released Mif acted through Cd74 receptor in HAM to promote immunochemotactic activity of microglia, triggering immuno-inflammatory responses in aged brains. These findings may provide new targets for reducing age-related brain inflammation and maintaining brain health.
