Cryo-EM structure of human glucose transporter GLUT4

Yafei Yuan et al.

Summary: In this study, the authors reveal the cryo-EM structure of human GLUT4 and compare it with GLUT1. They also lay the foundation for further investigation of GLUT4 trafficking and provide methods for cryo-EM analysis of other small solute carriers.

Methods of Liposomes Preparation: Formation and Control Factors of Versatile Nanocarriers for Biomedical and Nanomedicine Application

Domenico Lombardo et al.

Summary: Liposomes are nano-sized vesicles commonly used in nanomedicine and biomedical applications. They have high biocompatibility and can enhance drug delivery by reducing side effects and toxicity. However, liposomes are sensitive to mechanical and chemical actions, leading to limited colloidal stability and reduced cargo molecule entrapment.

The GFP thermal shift assay for screening ligand and lipid interactions to solute carrier transporters

Yurie Chatzikyriakidou et al.

Summary: Solute carrier (SLC) transporters, representing the second-largest fraction of the membrane proteome, have been underutilized as drug targets. The GFP thermal shift (GFP-TS) assay is a useful tool for detecting specific ligand interactions and affinities of SLC transporter fusions.

Structural comparison of GLUT1 to GLUT3 reveal transport regulation mechanism in sugar porter family

Tania Filipa Custodio et al.

Summary: The study compared the structures and functional data of GLUT1 and GLUT3, revealing significant differences in glucose affinity between them, possibly due to interactions between the cytosolic SP motif and conserved A motif. Additionally, previously undescribed Cl- ion site and endofacial lipid/glucose binding site in GLUT1 were identified as modulators of GLUT kinetics.

Structures and General Transport Mechanisms by the Major Facilitator Superfamily (MFS)

David Drew et al.

Summary: The major facilitator superfamily (MFS) is the largest known superfamily of secondary active transporters, responsible for transporting a wide range of substrates with remarkable diversity and complex gating mechanisms. Studies have shown that residues far from the substrate binding site in MFS transporters can be just as important, and some have evolved to form unique complexes. Through in-depth descriptions of glucose transporters and multidrug resistance antiporters, novel refinements to the rocker-switch alternating-access model have been proposed.

GLUT5 (SLC2A5) enables fructose-mediated proliferation independent of ketohexokinase

Roger J. Liang et al.

Summary: The ability of cells to proliferate in fructose is not determined by the tissue of origin or the expression level of specific genes related to fructose catabolism, but rather by chronic exposure to fructose. Upregulation of SLC2A5, encoding the fructose transporter GLUT5, enables cells to grow in fructose-containing media. Overexpression of GLUT5 allows fructose to be metabolized through glycolysis using hexokinase, indicating that fructose uptake is the key factor for fructose-mediated cell proliferation, regardless of ketohexokinase expression.
