Deciphering triterpenoid saponin biosynthesis by leveraging transcriptome response to methyl jasmonate elicitation in Saponaria vaccaria

Elucidation of the pathway for biosynthesis of saponin adjuvants from the soapbark tree

Summary: The Chilean soapbark tree produces QS saponins, which are important vaccine adjuvants. By sequencing the Q. saponaria genome, we identified 16 pathway enzymes and successfully produced advanced QS pathway intermediates in tobacco. We also identified the enzymes necessary for the production of QS-7, a saponin with excellent therapeutic properties. Our findings enable the production of Q. saponaria vaccine adjuvants and provide new avenues for developing immunostimulants.

The results suggested that the active components of PNS may exert therapeutic effects through apoptotic and cytokine signaling pathways. In addition, PNS was found to promote the proliferation of human umbilical vein endothelial cells (HUVECs) and angiogenesis.
