Actin Filaments Couple thes Protrusive Tips to the Nucleus through the I-BAR Domain Protein IRSp53 during the Migration of Cells on 1D Fibers

Summary: This study investigates the role of IRSp53 as a regulator of membrane curvature during cell migration on single, suspended 1D fibers. IRSp53 depletion reduces actin stress fibers, protrusive activity, and contractility, leading to uncoupling of the nucleus from cellular movements. The findings demonstrate the unique role of IRSp53 in controlling cell migration in 3D.

A minimal cell model for lamellipodia-based cellular dynamics and migration

Summary: A sheet-like protrusion called a lamellipodium plays a key role in cell motility. Recent theoretical studies have revealed a self-organizing feedback mechanism involving membrane-bound protein complexes and the cytoskeleton which can explain the formation of lamellipodia. This mechanism not only accounts for cell spreading and migration, but also explains processes such as phagocytosis. These findings suggest the presence of general physical principles underlying cellular shape dynamics.

Quantitative biophysical metrics for rapid evaluation of ovarian cancer metastatic potential

Summary: Ovarian cancer is often diagnosed at a late stage, and there are currently no reliable cellular markers to predict metastasis. In this study, researchers used simulated extracellular matrix fiber networks to study the progression and metastasis of ovarian cancer, and found that the degree of coiling at the tips of protrusions was strongly correlated with metastatic potential. This finding was confirmed in human ovarian cancer cell lines, providing a method to accurately assess ovarian cancer metastatic potential.

Sculpting Rupture-Free Nuclear Shapes in Fibrous Environments

Summary: This study investigates the plasticity of nucleus morphology in fibrous environments using suspended nanofiber networks. Contrary to cells on 2-dimensional surfaces, cells on fibers display a uniform actin network caging the nucleus, which plays a role in shaping nuclear shapes. The authors found that fiber diameter affects cell contractility and nuclear shape, and is also correlated with the translocation of mechanosensitive transcription factor Yes-associated protein (YAP) to the nucleus.

Schwann cell interactions during the development of the peripheral nervous system

Summary: Schwann cells are crucial in the development of the peripheral nervous system (PNS), establishing vital relationships with the extracellular environment and other cell types, particularly neurons. This review discusses the important interactions of Schwann cells in the proper establishment and function of the PNS, including signals from axons and the extracellular matrix, bidirectional signals to establish the axo-glial relationship, and how Schwann cells support axons. In addition, the speculation is made on how Schwann cell interactions with other components of the developing PNS ultimately promote the complete construction of the peripheral nerve.

The WAVE complex associates with sites of saddle membrane curvature

Summary: The study reveals that the WAVE complex forms actin-independent rings at regions of saddle membrane curvature, suggesting its role in organizing sheet-like lamellipodia and other emergent cell behaviors beyond Arp2/3 complex activation.
