Dynamic patterns of microRNA expression during acute myeloid leukemia state-transition

David E. Frankhouser et al.

Summary: This study analyzed time-series miRNA sequencing data in a mouse model to investigate the temporal dynamics of miRNA expression in AML. The results showed that the miRNA transcriptome undergoes state-transition during AML initiation and progression. By modeling AML state-transition, the researchers were able to accurately predict the time to AML and align samples from mice with different rates of AML development, providing a framework for studying the dynamic processes involved in AML.

Treatment of blast phase chronic myeloid leukaemia: A rare and challenging entity

Mhairi Copland

Summary: Despite the success of BCR-ABL-specific tyrosine kinase inhibitors (TKIs) in the treatment of chronic phase chronic myeloid leukemia (CML), blast phase CML (BP-CML) remains a challenging clinical need due to its rarity and heterogeneity. Further research and clinical trials are required to improve patient outcomes.

Mutations in the miR-142 gene are not common in myeloproliferative neoplasms

Paulina Galka-Marciniak et al.

Summary: Recent data shows that MIR142 is one of the most frequently mutated miRNA genes and noncoding elements in all cancers, especially in blood cancers. Mutations in MIR142 have profound consequences for lympho- and myelopoiesis, and one of its targeted genes, CD274, is elevated in many cancer types, including MPNs. However, MIR142 mutations are rare in MPNs.

Treatment-induced arteriolar revascularization and miR-126 enhancement in bone marrow niche protect leukemic stem cells in AML

Bin Zhang et al.

Summary: In FLT3-ITD+ AML models, treatment-induced changes in the BM vascular compartment impact the activity of leukemic stem cells (LSCs). TNF alpha mediates the loss of arterioles and gain of sinusoids in the BM niche by downregulating EC miR-126, affecting LSC growth. TKI treatment promotes re-vascularization and increases EC miR-126 supply to LSCs for protection against leukemia growth.
