Temporal analyses of postnatal liver development and maturation by single-cell transcriptomics

Yan Liang et al.

Summary: This study analyzed single-cell transcriptomes of mouse livers and identified hepatocyte heterogeneity and the zonated metabolic functions. It also discovered a group of macrophages with a hybrid phenotype that may play a role in sinusoidal construction and Treg-cell function. The comprehensive atlas provided by this study is important for understanding liver development, metabolism, and disease.

Follistatin-like 1 (FSTL1) interacts with Wnt ligands and Frizzled receptors to enhance Wnt/β-catenin signaling in obstructed kidneys in vivo

Yu Zhang et al.

Summary: The study reveals that FSTL1 plays a promoting role in renal fibrosis by interacting with Wnt ligands and FZD receptors to activate the Wnt/β-catenin pathway. This finding is of great importance for a deeper understanding of the mechanism underlying kidney diseases.

Obese Skeletal Muscle-Expressed Interferon Regulatory Factor 4 Transcriptionally Regulates Mitochondrial Branched-Chain Aminotransferase Reprogramming Metabolome

Ting Yao et al.

Summary: The study reveals that increased levels of interferon regulatory factor 4 (IRF4) in skeletal muscle are associated with obesity. Mice with specific IRF4 loss in skeletal muscle showed protection against the metabolic effects of high-fat diet. IRF4 regulates skeletal muscle metabolism through the upregulation of mitochondrial branched-chain aminotransferase (BCATm) expression.

A researcher's guide to preclinical mouse NASH models

Suchira Gallage et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) and its inflammatory form, non-alcoholic steatohepatitis (NASH), have become the most prevalent chronic liver diseases in the Western world and are risk factors for hepatocellular carcinoma (HCC). Despite the lack of clinically approved drugs for NAFLD, it is crucial to understand the pathogenesis of this disease and develop new therapies. Preclinical mouse models play a crucial role in investigating mechanisms and treatment options for NASH and subsequent HCC. However, not all models accurately replicate the human pathology. This Perspective highlights the advantages and disadvantages of different mouse models, with an emphasis on the use of 'Western diets'.

Myostatin as a fibroblast-activating factor impacts on postoperative outcome in patients with hepatocellular carcinoma

Sachiyo Yoshio et al.

Summary: High levels of serum myostatin are associated with poor prognosis in patients with non-alcoholic fatty liver disease-hepatocellular carcinoma (NAFLD-HCC) without advanced liver fibrosis. Myostatin activates primary liver fibroblasts and stimulates collagen production, indicating its potential role in disease progression.

Dynamic resistance exercise increases skeletal muscle-derived FSTL1 inducing cardiac angiogenesis via DIP2A-Smad2/3 in rats following myocardial infarction

Yue Xi et al.

Summary: The study found that dynamic resistance exercise can stimulate skeletal muscle to produce FSTL1 protein, promoting myocardial angiogenesis, inhibiting pathological remodeling, and protecting cardiac function. Exogenous FSTL1 can promote endothelial cell proliferation and regulate angiogenesis through the DIP2A-Smad2/3 signaling pathway.
