Molecular drivers of insecticide resistance in the Sahelo-Sudanian populations of a major malaria vector Anopheles coluzzii

Sulaiman S. Ibrahim et al.

Summary: Transcriptional analysis of Anopheles coluzzii populations from the Sahel region revealed overexpression of genes associated with insecticide resistance, including CYP450s, glutathione S-transferases, carboxylesterases, and cuticular proteins. High frequencies of known markers of resistance were also observed. These findings have important implications for malaria control efforts and regional collaboration.

Modelling spatiotemporal trends in the frequency of genetic mutations conferring insecticide target-site resistance in African mosquito malaria vector species

Penelope A. Hancock et al.

Summary: This study developed a statistical spatiotemporal model to investigate target-site resistance in malaria vectors in Africa. The results showed variations in the spread dynamics of resistance mutations across different vector species and geographic regions. The coverage of insecticide-treated bed nets was found to have varying effects on allele frequencies in different species and regions.

In vivo functional validation of the V402L voltage gated sodium channel mutation in the malaria vector An. gambiae

Jessica Williams et al.

Summary: The study identified the presence of the V402L-I1527T haplotype in resistant An. coluzzii strains and field populations from Burkina Faso, at frequencies higher than previously reported. Functional validation of V402L in insecticide resistance showed reduced mortality after exposure to pyrethroids and DDT, with lower levels compared to L995F. Unlike L995F, mosquitoes carrying V402L did not show any fitness costs under laboratory conditions.

Multi-omics analysis identifies a CYP9K1 haplotype conferring pyrethroid resistance in the malaria vector Anopheles funestus in East Africa

Jack Hearn et al.

Summary: This study investigates the molecular basis of metabolic resistance to pyrethroids in Anopheles funestus, a major malaria vector. The researchers used a multi-omics approach and functional validation to identify a cytochrome P450, CYP9K1, as a major driver of resistance. The study also identified other genes and variants associated with resistance and suggests their potential use in designing field-applicable markers for tracking resistance in Africa-wide.

Identification of a rapidly-spreading triple mutant for high-level metabolic insecticide resistance in Anopheles gambiae provides a real-time molecular diagnostic for antimalarial intervention deployment

Harun Njoroge et al.

Summary: Studies on insecticide resistance provide insights into the rapid evolutionary response of populations to contemporary selection. This study identified twin mutations and a triple-mutant haplotype that can predict mosquito resistance to pyrethroid insecticides, and found that the use of synergists in long lasting insecticidal nets can increase mortality in mosquitoes carrying the triple-mutant haplotype.

Resistance to pirimiphos-methyl in West African Anopheles is spreading via duplication and introgression of the Ace1 locus

Xavier Grau-Bove et al.

Summary: Research has shown that controlling mosquito populations is crucial for reducing malaria transmission in Africa. Resistance to the insecticide pirimiphos-methyl is primarily caused by mutations in a specific gene. These mutations are present in certain populations and have spread to other populations through inter-specific hybridization.

The genetic architecture of target-site resistance to pyrethroid insecticides in the African malaria vectors Anopheles gambiae and Anopheles coluzzii

Chris S. Clarkson et al.

Summary: Resistance to pyrethroid insecticides in malaria vectors is a major concern, with research showing widespread genetic variation in the Vgsc gene across African countries. New alleles were identified, some of which could enhance or compensate for known resistance genotypes. The study also revealed the potential for geographical spread of resistance through the identification of distinct haplotype groups carrying resistance alleles across multiple countries.

Integration of whole genome sequencing and transcriptomics reveals a complex picture of the reestablishment of insecticide resistance in the major malaria vector Anopheles coluzzii

Victoria A. Ingham et al.

Summary: Insecticide resistance poses a major threat to malaria control efforts, with studies showing that multiple changes in gene expression, genome, and microbiome contribute to the complex phenotype. Identifying these mechanisms is crucial for designing effective control strategies.

CRISPR/Cas9 modified An. gambiae carrying kdr mutation L1014F functionally validate its contribution in insecticide resistance and combined effect with metabolic enzymes

Linda Grigoraki et al.

Summary: The study demonstrates that introducing the 1014F allele in Anopheles mosquitoes significantly increases resistance to pyrethroids and DDT, while reducing mortality after exposure to pyrethroid-treated bednets. Combining 1014F with over-expression of detoxification enzymes further enhances resistance, although mosquitoes carrying the 1014F allele exhibit fitness disadvantages.

Threats to the effectiveness of insecticide-treated bednets for malaria control: thinking beyond insecticide resistance

Steve W. Lindsay et al.

Summary: Insecticide-treated bednets (ITNs) have been effective in reducing malaria morbidity and mortality in sub-Saharan Africa from 2004 to 2019, but the decline in malaria cases and deaths has stalled recently. It is important to consider factors beyond insecticide resistance, such as access to nets, use of nets, bioefficacy, and durability, in evaluating the effectiveness of ITNs. To improve the impact of ITNs, efforts should focus on increasing net coverage, funding for programmes, strategies to increase ITN uptake, and developing novel vector control tools.
