Chloramine-T-Enabled Mass Spectrometric Analysis of C=C Isomers of Unsaturated Fatty Acids and Phosphatidylcholines in Human Thyroids

Bo Zhang et al.

Summary: Specific locations of carbon-carbon double bonds (C=C) in lipids play important roles in biological processes. A N-tosylaziridination method was developed to annotate C=C positions in lipids using mass spectrometry, and it showed high efficiency, sensitivity, and specificity. Studies on human thyroid tissues suggested that increased unsaturated fatty acid content and n-6/n-3 ratios were closely associated with thyroid tumorigenesis.

Wenpeng Zhang et al.

Summary: In-depth structural characterization of lipids is crucial for lipidomics. Advances in mass spectrometry methods have allowed the resolution of lipid isomers at different structural levels, enabling the study of lipid metabolism and discovery of new biomarkers. This review focuses on recent developments in tandem mass spectrometry methods for identifying complex lipids beyond known headgroup and chain composition levels. The integration of isomer-resolving MS/MS methods with different lipid analysis workflows and their applications in lipidomics are also discussed.

The role of lipids in cancer progression and metastasis

Miguel Martin-Perez et al.

Summary: Lipids play essential roles in the body, but excessive lipids can promote tumor formation and metastasis. Cancer cells utilize lipid metabolism to adapt to the metastatic process and to resist treatment and promote relapse. Metabolic interventions aimed at reducing lipid availability to cancer cells provide promising opportunities for cancer prevention and treatment of metastasis.

Isomer Selective Comprehensive Lipidomics Analysis of Phosphoinositides in Biological Samples by Liquid Chromatography with Data Independent Acquisition Tandem Mass Spectrometry

Peng Li et al.

Summary: Phosphoinositides (PIPx) are crucial in cellular functions like growth, proliferation, differentiation, and are regulated by fatty acyl chains, degree of phosphorylation, and phosphorylation positions. However, there is a lack of comprehensive analytical method for distinguishing all regioisomeric forms with different acyl side chains and degrees of phosphorylation. This study presents a powerful method for analyzing PIPx profiles in biological samples using a chiral polysaccharide stationary phase and high-resolution mass spectrometry.
