Dual methylation and hydroxymethylation study of alcohol use disorder

Summary: This study characterized methylation and hydroxymethylation changes associated with alcohol use disorder (AUD) in blood and brain using an integrative, multi-tissue design. Cell-type-specific analyses in blood identified significant associations in monocytes and T-cells, with the monocyte findings significantly enriched for AUD-related methylation and hydroxymethylation in the brain. The study also discovered a novel mechanism for cognitive deficits associated with AUD and suggested promising new avenues for research in this area.

Intergenerational effects of preconception opioids on glucose homeostasis and hepatic transcription in adult male rats

Summary: Adolescence represents a critical period for neurodevelopment and adverse experiences during this time can have long-lasting effects on disease vulnerability and even impact future generations. This study demonstrates that male offspring of female rats exposed to morphine during adolescence are predisposed to the adverse effects of a high fat moderate sucrose diet, displaying increased levels of fasting glucose and insulin. RNA sequencing revealed significant differences in the hepatic transcriptome, suggesting alterations in metabolism. These findings highlight the intergenerational effects of adolescent opioid exposure on the risk of developing insulin resistance and associated comorbidities.

Summary: Despite the significant impact of opioid use disorder (OUD), previous genome-wide association studies (GWAS) of OUD have identified a limited number of susceptibility loci. In this large-scale GWAS study, researchers optimized genetic informativeness by performing multi-trait analysis of GWAS (MTAG) with genetically correlated substance use disorders (SUDs) in individuals of European and African ancestry. The study identified several genetic associations with OUD, including variants in the OPRM1 gene and a variant in the FURIN gene, as well as 18 genome-wide significant loci associated with OUD-MTAG.
