Phagocytosis and self-destruction break down dendrites of Drosophila sensory neurons at distinct steps of Wallerian degeneration

Hui Ji et al.

Summary: After injury, dendrites and axons expose phosphatidylserine (PS) and self-destruct. The degeneration of injured axons is controlled by a conserved pathway, activating neurite self-destruction through NAD+ depletion. Phagocytosis is the main driver of dendrite degeneration induced by both genetic disruptions and injury. Both PS exposure and self-destruction contribute to dendrite degeneration in injury, with Axed acting alongside phagocytosis. Injured dendrites exhibit rhythmic calcium-flashing correlated with degeneration.

Delta/Notch signaling in glia maintains motor nerve barrier function and synaptic transmission by controlling matrix metalloproteinase expression

Mario R. Calderon et al.

Summary: This study reveals that Delta/Notch signaling in subperineurial glia plays an essential role in controlling the expression of matrix metalloproteinase 1 (Mmp1), which is crucial for the extracellular matrix (ECM). In the absence of this signaling, enhanced Mmp1 activity disrupts septate junctions and glial ensheathment, leading to compromised neurotransmitter release.

Structural and Functional Synaptic Plasticity Induced by Convergent Synapse Loss in the Drosophila Neuromuscular Circuit

Yupu Wang et al.

Summary: In Drosophila larval neuromuscular junctions, ablating specific types of motor neurons can lead to structural changes and increased neurotransmitter release at other motor neuron synapses, demonstrating synaptic plasticity. Some synapses exhibit compensatory plasticity, while silencing one type of motor neuron can affect the plasticity of other types of synapses.

Astrocytes phagocytose adult hippocampal synapses for circuit homeostasis

Joon-Hyuk Lee et al.

Summary: This study demonstrates that astrocytes in the adult hippocampus carry out phagocytosis through MEGF10 to maintain proper synaptic connectivity and plasticity. The findings reveal the crucial role of astrocytes in the activity-dependent elimination of excitatory synapses, highlighting the importance of this astrocytic function in supporting cognitive function.

Injury-Induced Inhibition of Bystander Neurons Requires dSarm and Signaling from Glia

Jiun-Min Hsu et al.

Summary: This study reveals the effects of nervous system injury on functional connectivity and finds that axotomy leads to impaired axon transport and reduced signal transduction in bystander neurons. The research also identifies a critical role for dSarm and glia in signaling and functional recovery in the injured nervous system.

Astrocytes close a motor circuit critical period

Sarah D. Ackerman et al.

Summary: The study identifies a critical period in a developing Drosophila motor circuit and demonstrates the essential role of astrocytes in proper critical period termination. Changes in activity during the critical period regulate dendrite length, complexity, and connectivity of motor neurons, while astrocyte ablation prolongs this critical period. Additionally, genetic screening reveals astrocyte-motor neuron signaling pathways that ensure timely critical period closure and proper locomotor behavior.
