Abnormal dynamic functional connectivity during fear extinction learning in PTSD and anxiety disorders

Zhenfu Wen et al.

Summary: Research on fear/threat extinction has shown that individuals with anxiety disorders (AX) or posttraumatic stress disorder (PTSD) demonstrate widespread abnormal functional connectivity in higher-order cognitive and attention networks during extinction learning, especially involving connections related to the dorsolateral prefrontal cortex. Furthermore, impaired dynamic functional connectivity during extinction learning is correlated with fear and anxiety-related clinical measures.

Sex differences in the immediate extinction deficit and renewal of extinguished fear in rats

Annalise N. Binette et al.

Summary: Extinction learning is crucial in reducing fear and anxiety, but patients with fear and anxiety disorders are often resistant to this type of learning. Additionally, trauma and stress-related disorders tend to relapse more in females compared to males, suggesting females may be more susceptible to extinction deficits and fear relapse. This study examined sex differences in a stress-induced extinction learning impairment and fear relapse in rats. The results showed that while there were no sex differences in the immediate extinction deficit, male rats exhibited significantly greater fear relapse compared to females.

Neural Oscillations in Aversively Motivated Behavior

Michael S. Totty et al.

Summary: This article reviews the oscillatory correlates of fear and anxiety, with a focus on theta and gamma rhythms. The study finds that hippocampal network oscillations play a role in integrating spatial and motivational information during anxiety states, and these oscillations also synchronize different brain structures to guide adaptive behavior. Additionally, theta and gamma oscillations are important in the encoding and retrieval of fear memories.

Coupling between slow waves and sharp-wave ripples engages distributed neural activity during sleep in humans

Ivan Skelin et al.

Summary: Studies suggest that during sleep, hippocampal sharp-wave ripples and other neural activities can modulate widespread high-frequency activity, predicting coordinated activity between different brain regions. These findings imply a mechanism where hippocampal and cortical slow-wave synchronization supports memory consolidation.

L-DOPA modulates activity in the vmPFC, nucleus accumbens, and VTA during threat extinction learning in humans

Roland Esser et al.

Summary: Learning to be safe is crucial for adaptive behavior, while detecting the absence of expected threats is key for threat extinction learning. A possible mechanism for extinction learning involves a dopaminergic mismatch signal, which encodes the absence of expected threats.
