Deficiency of immune-responsive gene 1 exacerbates interleukin-1beta-elicited the inflammatory response of chondrocytes via enhancing the activation of NLRP3 inflammasome

Summary: This study found that immune-responsive gene 1 (IRG1) plays an important role in osteoarthritis and is related to the inflammatory response of chondrocytes. The expression of IRG1 increases after stimulation with IL-1 beta, and its deficiency exacerbates IL-1 beta-induced apoptosis and degradation of the extracellular matrix in chondrocytes. IRG1 deficiency also enhances NLRP3 inflammasome activation induced by IL-1 beta. The supplementation of itaconate can improve the inflammatory response induced by IL-1 beta and reverse the effects of IRG1 deficiency.
