The key role of proteostasis at mitochondria-associated endoplasmic reticulum membrane in vanadium-induced nephrotoxicity using a proteomic strategy

Summary: Excessive vanadium contamination is a growing concern due to its negative effects on human health and ecosystems, particularly nephrotoxicity. The role of mitochondria-associated endoplasmic reticulum membrane (MAM) in vanadium-induced nephrotoxicity has been investigated. Proteomics analysis revealed that vanadium primarily affects MAM-mediated metabolic pathways, particularly the endoplasmic reticulum (ER) proteostasis pathway. These findings provide promising insights into the toxicological mechanisms of vanadium and the critical role of MAM.

Summary: The enzyme Acox2 is involved in bile acid synthesis and fatty acid degradation. Knockout mice for Acox2 spontaneously developed liver cancer with lymphocytic infiltrate. The study found that non-histone lysine crotonylation (Kcr) levels were decreased in the livers of Acox2(-/-) mice. The Kcr signals were concentrated in the nucleus of tumor cells but mostly located in the cytoplasm of normal liver cells. Further analysis revealed that downregulated Kcr sites were mainly located in mitochondrial and peroxisomal enzymes. Mutagenesis and analysis showed that a specific Kcr site might regulate TOP3B expression, leading to increased DNA damage.
