Targeting KDM4C enhances CD8+ T cell mediated antitumor immunity by activating chemokine CXCL10 transcription in lung cancer

Xiaohua Jie et al.

Summary: The study reveals that inhibition of KDM4C can increase CD8(+) T cell infiltration, promote the proliferation, migration and activation of CD8(+) T cells, and alleviate CD8(+) T cell exhaustion in mouse tumor tissues. Mechanistically, KDM4C inhibition increases the binding of H3K36me3 to the CXCL10 promoter region, thus inducing CXCL10 transcription and enhancing the CD8(+) T cell mediated antitumor immune response. Among the tested regimens, the triple therapy achieved the best therapeutic efficacy with tolerable toxicity in lung cancer.

The Emerging Significance of Histone Lysine Demethylases as Prognostic Markers and Therapeutic Targets in Head and Neck Cancers

Dawid Dorna et al.

Summary: Epigenetic aberrations, including altered DNA methylation and histone modifications, are commonly found in head and neck squamous cell carcinomas (HNSCC). Histone lysine demethylases (KDMs) have been implicated in the pathogenesis of HNSCC and are potential therapeutic targets. These enzymes play a role in transcriptional regulation and various biological processes. Overexpression of KDMs in HNSCC affects cell proliferation, apoptosis, cell motility, and stemness.

SMYD5 catalyzes histone H3 lysine 36 trimethylation at promoters

Yanjun Zhang et al.

Summary: SETD2 methylates histone H3K36me3 in gene bodies in mammalian cells. In this study, the authors demonstrate that H3K36me3 is also enriched at promoter regions and is catalyzed by SMYD5, recruited by RNA polymerase II. They further show that SMYD5 is elevated in liver cancer, with a correlation to changes in gene expression. These findings provide insights into the localization and function of H3K36me3.

Mechanistic insights into KDM4A driven genomic instability

Nicolas L. Young et al.

Summary: Alterations in global epigenetic signatures on chromatin, specifically the overexpression of demethylase KDM4A, play a crucial role in tumor initiation and progression by modulating key cellular processes such as transcription and proliferation. This highlights the therapeutic potential of targeting KDM4A and other demethylases in cancer treatment.

LEF1 silencing sensitizes colorectal cancer cells to oxaliplatin, 5-FU, and irinotecan

Elham Fakhr et al.

Summary: LEF1 plays an important role in the chemo-resistance of colorectal cancer cells, with downregulation of the long isoform making cells more sensitive to chemotherapy through reduced proliferation, increased apoptosis, or cell cycle arrest. Balancing the expression of long and short isoforms of LEF1 is crucial in overcoming chemo-resistance.

The inhibitors of KDM4 and KDM6 histone lysine demethylases enhance the anti-growth effects of erlotinib and HS-173 in head and neck cancer cells

Robert Kleszcz et al.

Summary: This study investigated the anti-cancer effects of KDM4 and KDM6 inhibitors, both alone and in combination with EGFR and PI3K inhibitors, on HNSCC cells. The results showed that these compounds and their combinations induced apoptosis and were associated with changes in gene expression levels, suggesting that inhibition of KDM4 and KDM6 could be a novel therapeutic strategy in HNSCC.

H3K36 trimethylation-mediated biological functions in cancer

Chu Xiao et al.

Summary: Histone H3 lysine 36 trimethylation (H3K36me3) plays a crucial role in genome regulation, especially in cancer pathogenesis. Disruption of the histone methylation regulatory network and mutations in H3K36me3 codes may have a powerful impact on cancer initiation and development. Understanding the role of H3K36me3 in cancer can lead to targeted therapeutic strategies for cancer treatment.

Targeting KDM4A epigenetically activates tumor-cell-intrinsic immunity by inducing DNA replication stress

Wuchang Zhang et al.

Summary: Inhibition of KDM4A promotes heterochromatin compaction and DNA replication stress in SCC, activating tumor-cell-intrinsic cGAS-STING signaling. KDM4A inhibition collaborates with PD1 blockade to inhibit SCC growth and metastasis, efficiently eliminating cancer stem cells. Targeting KDM4A can activate anti-tumor immunity and enhance PD1 blockade immunotherapy in SCC by aggravating replication stress.

PRMT6 activates cyclin D1 expression in conjunction with the transcription factor LEF1

Lucas Schneider et al.

Summary: PRMT6 is an epigenetic regulator that influences cellular differentiation and proliferation through methylating arginines at histone H3. Transcription factors such as LEF1 are recruited by PRMT6 to regulate gene expression, particularly for key cell cycle related target genes like CCND1. Knockdown of LEF1 or PRMT6 leads to reduced CCND1 expression, affecting cell cycle progression and proliferation. These findings enhance understanding of PRMT6 activity in cell cycle regulation and suggest potential for developing specific PRMT6 inhibitors for cancer therapy.

Summary: The study revealed that ASPN is upregulated in gastric cancer and associated with poor prognosis. Oncogenic PAX6 elicits CDK4/6 inhibitor resistance by epigenetically inactivating the LATS2-Hippo signaling pathway

Yi Zhang et al.

Summary: PAX6 expression negatively correlates with response to palbociclib in gastric cancer by hypermethylating the promoter of LATS2 and inactivating the Hippo pathway, resulting in upregulation of cyclin D1 and resistance to palbociclib. Activation of the Hippo signaling pathway or treatment with a DNA methylation inhibitor can overcome PAX6-induced resistance to palbociclib in gastric cancer.
