Acetyl-CoA biosynthesis drives resistance to histone acetyltransferase inhibition

Summary: Histone acetyltransferases (HATs) are involved in both oncogene and nononcogene dependencies in human cancers. Acetyl-CoA-competitive HAT inhibitors have potential as cancer therapeutics, but understanding of therapeutic response and drug resistance is limited. This study reveals that elevated acetyl-CoA concentrations can cause acquired drug resistance by outcompeting drug-target engagement. This finding applies not only to CBP/p300 HAT inhibitors, but also to an investigational KAT6A/B HAT inhibitor currently in Phase 1 clinical trials.
