UniProt: the Universal Protein Knowledgebase in 2023

Alex Bateman et al.

Summary: The UniProt Knowledgebase aims to provide comprehensive, high-quality, and freely accessible protein sequences annotated with functional information. The database has expanded its data processing pipeline and website to accommodate the increasing information content, with over 227 million sequences and plans to include a reference proteome for each taxonomic group. Detailed annotations are extracted from the literature to update or create reviewed entries, while unreviewed entries are supplemented with annotations from automated systems. The new website, https://www.uniprot.org/, offers enhanced user experience and easy access to data, including AlphaFold structures and improved protein subcellular localization visualizations.

Fucosylation of HLA-DRB1 regulates CD4+ T cell-mediated anti-melanoma immunity and enhances immunotherapy efficacy

Daniel K. Lester et al.

Summary: Immunotherapy efficacy in melanoma is limited, and combinations with other therapies have shown limited improvements but also adverse events. The lack of intratumoral immune cells (itICs) further impairs efficacy, requiring strategies to safely increase itICs. This study shows that dietary administration of l-fucose induces fucosylation and cell surface enrichment of HLA-DRB1 in melanoma cells, leading to increased itICs and anti-tumor immunity. This suggests that l-fucose could be a powerful agent for enhancing immunotherapy efficacy in melanoma.

Unconventional protein post-translational modifications: the helmsmen in breast cancer

Jiena Liu et al.

Summary: Breast cancer is the most common malignant tumor in women worldwide, and post-translational modifications (PTMs) of proteins play important roles in tumorigenesis and progression. However, the study of non-histone modifications, particularly those other than phosphorylation, has been relatively limited. This review aims to explore the functions and impact of these PTMs in breast cancer, and emphasizes the importance of developing novel therapeutic drugs targeting PTMs and biological markers for breast cancer detection.

ESRP1-regulated isoform switching of LRRFIP2 determines metastasis of gastric cancer

Jihee Lee et al.

Summary: The epithelial and mesenchymal isoform switches of LRRFIP2, regulated by ESRP1, are closely related to the metastatic potential of gastric cancer cells. The epithelial isoform of LRRFIP2 inhibits the oncogenic function of CARM1, while the mesenchymal isoform promotes liver metastasis.

The Histone Methyltransferase SETD8 Regulates the Expression of Tumor Suppressor Genes via H4K20 Methylation and the p53 Signaling Pathway in Endometrial Cancer Cells

Asako Kukita et al.

Summary: The histone methyltransferase SETD8, which methylates histone H4 lysine 20 (H4K20) and non-histone proteins such as p53, plays a critical role in the proliferation and apoptosis of endometrial cancer cells. SETD8 regulates genes through H4K20 methylation and the p53 signaling pathway, and its expression is elevated in endometrial cancer tissues. Additionally, prognostically important genes related to apoptosis have been identified.

JNK Mediates Differentiation, Cell Polarity and Apoptosis During Amphioxus Development by Regulating Actin Cytoskeleton Dynamics and ERK Signalling

Ildiko M. L. Somorjai et al.

Summary: The study demonstrates that pharmacological inhibition of JNK signaling in cephalochordate embryos results in morphological abnormalities such as notochord elongation defects, cell delamination, and altered gene expression. This suggests a conserved role of JNK signaling in regulating cellular behavior and tissue differentiation during cephalochordate morphogenesis.

Testicular orphan receptor 4 (TR4) promotes papillary thyroid cancer invasion via activating circ-FNLA/miR-149-5p/MMP9 signaling

Xiwu Ouyang et al.

Summary: The study highlights the oncogenic role of TR4 in the development of papillary thyroid cancer (PTC), driving PTC progression by promoting cell invasion and migration. The TR4/circFLNA/miR-149-5p/MMP9 signaling axis plays a central role in cell invasion and migration of PTC cells, suggesting that targeting this pathway may be an alternative option for metastatic PTC patients.
