Lassa fever - the road ahead

Robert F. Garry

Summary: Lassa virus is a locally endemic virus in West Africa that causes Lassa fever in humans through transmission from rodent reservoirs. Despite gaps in knowledge, efforts to develop countermeasures have increased due to frequent outbreaks and the potential risks posed by the virus.

Inhibition of cellular RNA methyltransferase abrogates influenza virus capping and replication

Yuta Tsukamoto et al.

Summary: Scientists have discovered the importance of host RNA modification in the initiation of influenza virus replication. They found that a derivative of a natural product called TFMT can inhibit this modification and restrict influenza virus replication. TFMT acts synergistically with approved anti-influenza drugs.

Pharmacological suppression of glycogen synthase kinase-3 reactivates HIV-1 from latency via activating Wnt/β-catenin/TCF1 axis in CD4+ T cells

Jing Wen et al.

Summary: In this study, the researchers identified the Wnt downstream beta-catenin/TCF1 pathway as a crucial modulator for HIV-1 latency. Activation of this pathway with GSK3 inhibitors effectively promoted HIV-1 proviral DNA transcription and reactivated latency in CD4(+) T cells.

β-catenin regulates HIV latency and modulates HIV reactivation

Hannah J. Barbian et al.

Summary: Latency is the main obstacle towards an HIV cure, and targeting multiple latency maintenance pathways is essential. In this study, beta-catenin, a transcriptional co-activator and central mediator of Wnt signaling, is identified as a regulator of HIV latency. Inhibiting or activating the beta-catenin pathway can reactivate or repress HIV, and it also enhances or inhibits the activity of other HIV latency reversing agents.

Recent advances of fl-catenin small molecule inhibitors for cancer therapy: Current development and future perspectives

Panpan Yang et al.

Summary: The Wnt/fl-catenin signaling pathway is involved in various cellular processes and its abnormal activation has been linked to cancer development. Fl-catenin, a core component of this pathway, is a key factor in its activation. Therefore, the development of Fl-catenin inhibitors has become an important focus in anticancer drug research. However, challenges in terms of selectivity, specificity, and physicochemical properties need to be addressed. This review highlights the progress made in the research of small molecule inhibitors targeting Fl-catenin and discusses the future prospects in drug development.

Structural insights into RNA polymerases of negative-sense RNA viruses

Aartjan J. W. te Velthuis et al.

Summary: RNA viruses, including important human and animal pathogens, have genomes consisting of single or multiple RNA segments. Replication and transcription are performed by RNA-dependent RNA polymerases, and recent high-resolution studies have provided insights into the structures of these enzymes and their roles in viral transcription and replication, offering potential targets for antiviral drug design.

β-Catenin Restricts Zika Virus Internalization by Downregulating Axl

Oscar A. Jimenez et al.

Summary: This study investigates the mechanisms of Zika virus entry into astrocytes, showing that the interaction between the Axl receptor and beta-catenin plays a crucial role in regulating viral internalization. The results highlight a dynamic host-virus interaction, where Zika virus downregulates beta-catenin to promote its internalization into susceptible target cells through the induction of Axl.
