Application of recyclable CRISPR/Cas9 tools for targeted genome editing in the postharvest pathogenic fungi Penicillium digitatum and Penicillium expansum

Summary: In this study, we successfully developed a method for implementing CRISPR/Cas9 technology in Penicillium digitatum and Penicillium expansum fungi for the first time, targeting the wetA gene. Mutants showed altered growth, pathogenicity, conidiogenesis, germination, and sensitivity to antifungal proteins. This work demonstrates the feasibility of the CRISPR/Cas9 system and expands the genetic engineering tools available for these postharvest pathogens.

Summary: In this study, the interaction between GalX, GalR, and AraR in the catabolism of d-galactose and l-arabinose was investigated in filamentous fungi. The results clearly demonstrated that GalX, GalR, and AraR co-regulate the catabolism of d-galactose in Aspergillus nidulans, with GalX playing a prominent role in gene regulation and AraR compensating for the absence of GalR and/or GalX.

Unraveling the regulation of sugar beet pulp utilization in the industrially relevant fungus Aspergillus niger

Evasion of Cas9 toxicity to develop an efficient genome editing system and its application to increase ethanol yield in Fusarium venenatum TB01

Summary: The CRISPR/Cas9 system is a powerful genome editing tool, but the toxicity of Cas9 limits its further application. In this study, the toxicity of Cas9 was eliminated and two more efficient methods for sgRNA expression were found for Fusarium venenatum TB01. By deleting the FvBDH gene, the ethanol yield was increased. This highly efficient CRISPR/Cas9 system lays the technical foundation for the metabolic engineering of F. venenatum TB01.

Occurrence and Nature of Off-Target Modifications by CRISPR-Cas Genome Editing in Plants

Summary: CRISPR-Cas-based genome editing allows for precise and targeted genetic modification of plants, but unintended off-target edits can increase risks for gene-edited food crops. Most observed off-target changes are small insertions, deletions, or nucleotide substitutions, with large deletions being rare. Off-target sites have few mismatches with the target sequence and are mainly located in protein-coding regions, often in target gene homologues. Off-target edits are primarily detected through biased analysis of predicted off-target sites. CRISPR-Cas-edited plants show lower off-target mutation frequencies compared to conventionally bred plants. This review can assist in discussing the relevance of evaluating off-target modifications for risk assessment of CRISPR-Cas-edited plants.
