Reversal of G-Quadruplexes' Role in Translation Control When Present in the Context of an IRES

Mohammed Enamul Hoque et al.

Summary: G-quadruplexes (GQs) are secondary nucleic acid structures that regulate cellular processes. They can both repress and initiate translation in mRNA's 5' UTR. Some mRNA's with GQs have been identified as positive regulators of translation. Cap-independent translation occurs through internal ribosomal entry sites (IRESs) in the 5' UTR.

uORF-introducing variants in the 5′UTR of the NIPBL gene as a cause of Cornelia de Lange syndrome

Juliette Coursimault et al.

Summary: Cornelia de Lange syndrome (CdLS) is a rare developmental disorder. This study identified variants in the 5'-untranslated region (UTR) of the NIPBL gene that may introduce an upstream open reading frame (uORF), leading to loss of function. The results suggest that uORF-introducing NIPBL variants may be a rare source of pathogenic variants in unsolved CdLS patients.

IRES-mediated Wnt2 translation in apoptotic neurons triggers astrocyte dedifferentiation

Hong Fan et al.

Summary: Reactive astrogliosis is enhanced by the activation of Wnt signaling and dedifferentiation of astrocytes following ischemic injury. Apoptotic neurons up-regulate Wnt2 protein via internal ribosome entry site (IRES)-mediated translation, leading to the activation of Wnt signaling and astrocyte dedifferentiation. This apoptosis-initiated Wnt-activating mechanism promotes neuronal regeneration.

Translational control in aging and neurodegeneration

Geena Skariah et al.

Summary: Protein metabolism plays crucial roles in aging and neurodegeneration, with changes in protein synthesis efficiency and accuracy less understood than degradation. Interventions that prolong lifespan often involve preemptively decreasing protein synthesis rates to harmonize with age-related declines in catabolism. Emerging technologies are revealing mRNA-specific changes in translation rates across tissues, pointing to key interactions between catabolic and anabolic pathways that contribute to functional decline.

Assessing eukaryotic initiation factor 4F subunit essentiality by CRISPR-induced gene ablation in the mouse

Patrick Senechal et al.

Summary: The translation initiation factor eIF4F is crucial for cap-dependent protein synthesis initiation. Different subunits of eIF4F have varying roles in development and tumorigenesis, with some being essential for viability and tumor onset, while others have non-essential functions. This study highlights the complex roles of different eIF4F subunits in gene expression regulation during embryogenesis and tumor development.
