Development of in ovo-compatible NS1-truncated live attenuated influenza vaccines by modulation of hemagglutinin cleavage and polymerase acidic X frameshifting sites

Summary: To develop more broadly protective vaccines for poultry production, modifications were made to a high interferon-inducing live attenuated influenza vaccine candidate. By replacing certain genetic sites in the vaccine virus, the pathogenicity was reduced for chicken embryos while maintaining protective efficacy. In ovo vaccination with the modified vaccine showed minimal effects on hatchability and protected chickens against a heterologous challenge virus.

These DIPs interfere with the replication of standard virus (STV) through competitive inhibition, making them potential antivirals for influenza treatment. By studying the propagation competition between different defective interfering (DI) vRNAs in an STV coinfection scenario, researchers have identified certain DI vRNAs with replication advantages and optimal fragment lengths that can be selected as promising candidates for efficacious antiviral therapy.
