Identification of Polyphenol Derivatives as Novel SARS-CoV-2 and DENV Non-Nucleoside RdRp Inhibitors

Summary: DF-47 and DF-51 were identified as effective inhibitors of SARS-CoV-2/DENV polymerase through RdRp inhibition screening and in vitro antiviral study. In silico simulation revealed stable binding modes between DF-47/DF-51 and SARS-CoV-2/DENV RdRp, including chelating with Mg2+ near the polymerase active site. These polyphenols have the potential to be developed into broad-spectrum, non-nucleoside RdRp inhibitors with a new scaffold.

A phytochemical library consisting of 2431 compounds from 104 Korean medicinal plants was screened using molecular docking and deep learning, leading to the identification of two lead compounds, Catechin gallate and Quercetin 3-O-malonylglucoside, with inhibitory potency against Mpro.

Summary: In this study, existing and novel non-nucleoside inhibitors for SARS-CoV-2 RdRp were computationally screened from large chemical databases. These inhibitors showed strong binding affinity with RdRp, drug-like features, good absorption, distribution, metabolism, and excretion profile, and were found to be non-toxic. These findings hold promise for the discovery of novel drugs against COVID-19 in the future.

The electrophile (warhead) is a crucial factor for covalent inhibitors, dictating selectivity and reactivity, and can be modified through rational designs. Covalent inhibitors are also finding applications in protein degradation and targeting chimeras (PROTACs) for 'undruggable' proteins, as well as in the treatment of SARS-CoV-2.

Further studies are needed to explore the action mechanisms of these protease inhibitors in controlling SARS-CoV-2 replication in human cells.
