Adeno-associated virus (AAV) cell entry: structural insights

Nancy L. Meyer et al.

Summary: Adeno-associated virus (AAV) is a leading vector for gene therapy of inherited diseases. The interactions between virus and host cells are complex and serotype-specific. Cryogenic electron microscopy (cryo-EM) provides a way to understand the structural details of virus-host complexes and uncover the steps of viral entry. Comparisons with antibody complexes also provide insights for improving immune neutralization.

Capsid Structure of Aleutian Mink Disease Virus and Human Parvovirus 4: New Faces in the Parvovirus Family Portrait

Renuk Lakshmanan et al.

Summary: This study determined the capsid structures of Aleutian mink disease virus (AMDV) and human parvovirus 4 (PARV4) using cryo-electron microscopy. Despite low amino acid sequence similarities, both viruses have icosahedral capsid structures and display major structural variabilities in surface loops.

pH-Induced Conformational Changes of Human Bocavirus Capsids

Mengxiao Luo et al.

Summary: Human bocaviruses (HBoVs) infect children and immunocompromised individuals, resulting in respiratory and gastrointestinal infections. This study compared the capsid structures of HBoV2 with previously determined HBoV1, HBoV3, and HBoV4, revealing strain and genus-specific features contributing to tissue tropism and antigenicity. Additionally, analyzing HBoV1 and HBoV2 structures under acidic conditions provided insights into capsid changes associated with endosomal and gastrointestinal acidification, unraveling the molecular mechanism of HBoV infection.

Completion of the AAV Structural Atlas: Serotype Capsid Structures Reveals Clade-Specific Features

Mario Mietzsch et al.

Summary: This study reports the capsid structures of four additional AAV serotypes, completing the structural atlas of AAV capsids. These structures exhibit AAV capsid features and show classification-specific protein loop conformations. The comprehensive characterization of these structures provides valuable insights for future vector engineering efforts to improve gene delivery success.

Characterization of the GBoV1 Capsid and Its Antibody Interactions

Jennifer Chun Yu et al.

Summary: The study reports the capsid structure of Gorilla bocavirus 1 (GBoV1) and its interactions with human sera and monoclonal antibodies. GBoV1 shares similarities with Human bocavirus 1 (HBoV1) but shows differences in specific structural regions. Furthermore, GBoV1 demonstrates comparable seropositivity rates to HBoV1 when tested against human sera.

Recent Advances in Molecular Biology of Human Bocavirus 1 and Its Applications

Liting Shao et al.

Summary: Human bocavirus 1 (HBoV1) is an autonomous human parvovirus that causes acute respiratory tract infections in children and infects human airway epithelial cells in vitro. It expresses multiple proteins and RNA, playing important roles in viral gene expression and replication, and serving as a helper virus for other viruses in human cells.
