Comparative cytotoxicity, endocrine-disrupting effects, oxidative stress of halophenolic disinfection byproducts and the underlying molecular mechanisms revealed by transcriptome analysis

Xiuwen Li et al.

Summary: Halophenolic disinfection byproducts (DBPs) are emerging pollutants that have adverse effects on human cells and need further exploration of molecular mechanisms. Substituting halophenolic DBPs with the same halogen leads to greater cytotoxicity when there are more substitution sites. Iodophenols are the most toxic, followed by bromo-phenols and chlorophenols when substituted at the same sites. Some of these DBPs showed significant endocrine-disrupting effects at sublethal concentrations.

Dimensionless Groups by Entropic Similarity: I - Diffusion, Chemical Reaction and Dispersion Processes

Robert K. K. Niven

Summary: Since Buckingham proposed dimensional analysis and similarity arguments based on dimensionless groups in 1914, they have been widely used in various branches of science and engineering. In this study, an additional category of dimensionless groups based on entropic similarity is proposed, which is defined by comparing ratios of entropy production terms, entropy flow rates or fluxes, and information flow rates or fluxes. The analysis expands the scope of dimensional analysis and similarity arguments for resolving new and existing problems in science and engineering by directly analyzing processes involving friction, dissipation, diffusion, dispersion, mixing, separation, chemical reaction, gain of information, and other irreversible changes in terms of competing entropy-producing and transporting phenomena and the dominant entropic regime, rather than indirectly in terms of forces.

Comparative Toxicity Analyses from Different Endpoints: Are New Cyclic Disinfection Byproducts (DBPs) More Toxic than Common Aliphatic DBPs?

Yun Wu et al.

Summary: The study compared the in vitro and in vivo toxicity of cyclic DBPs and aliphatic DBPs, revealing different toxicity rank orders with application of different endpoints. A Pearson correlation analysis of DBP toxicity data showed high correlation between observed cytotoxicity in Hep G2 cells and zebrafish embryo toxicity with previously reported in vitro and in vivo toxicity. The study also established two QSAR models to interpret the toxicity mechanisms of DBPs from different endpoints.

Formation and Cytotoxicity of Halophenylacetamides: A New Group of Nitrogenous Aromatic Halogenated Disinfection Byproducts in Drinking Water

Shaoyang Hu et al.

Summary: This study successfully identified a new group of nitrogenous aromatic halogenated disinfection byproducts, halophenylacetamides (HPAcAms), in laboratory experiments and realistic drinking water for the first time. The study investigated the formation mechanism, occurrence frequencies, and concentrations of HPAcAms in authentic drinking water, and developed a quantitative structure-activity relationship (QSAR) model based on cytotoxicity data. The results showed that HPAcAms could form from phenylalanine during chlorination and their yields were influenced by various factors. Multiple HPAcAms were detected in authentic drinking water samples and showed higher cytotoxicity compared to the most abundant aliphatic haloacetamide DBP.

Identification, Occurrence, and Cytotoxicity of Haloanilines: A New Class of Aromatic Nitrogenous Disinfection Byproducts in Chloraminated and Chlorinated Drinking Water

Di Zhang et al.

Summary: This study reports the presence of six new aromatic nitrogenous disinfection byproducts (DBPs) in drinking water. Haloanilines completely degrade in the presence of chlorine within 1 hour, but about 20% remains in the presence of chloramine after 120 hours. Haloanilines show high stability in the absence of disinfectants at pH 5-9. Eight haloanilines were detected in chloraminated finished water and tap water, with the highest total concentration reaching 443 ng/L. The cytotoxicity of haloanilines was lower than that of regulated DBPs. The formation and control of haloanilines in drinking water call for further investigation.

Halonaphthoquinones: A group of emerging disinfection byproducts of high toxicity in drinking water

Hangcheng Jiang et al.

Summary: Aromatic halogenated disinfection byproducts (DBPs) have high toxicity and have been the focus of research, but most studies have focused on halo-monocyclic DBPs, while halo-polycyclic DBPs have been largely unexplored. This study systematically studied a new group of halo-bicyclic DBPs called halonaphthoquinones (HNQs), and developed a highly accurate and sensitive method to detect HNQs in drinking water.

Halohydroxybenzonitriles as a new group of halogenated aromatic DBPs in drinking water: Are they of comparable risk to halonitrophenols?

Shaoyang Hu et al.

Summary: A new group of halogenated aromatic disinfection byproducts (DBPs), called halohydroxybenzonitriles (HHBNs), were detected in drinking water samples. These HHBNs exhibited high cytotoxicity and showed transformation during chlorination. Compared to regulated DBPs, HHBNs had similar concentration-cytotoxicity contribution and slightly weaker stability, making them a concern in drinking water.

Summary: Mitoxantrone, a well-known anthraquinone anti-cancer drug, has analogs 1PAQ and 2PAQ synthesized for studying their bioactivities. The analogs showed pharmacological properties comparable to Mitoxantrone, with 1-substituted analog exhibiting higher activity against Hep2C and MCF-7 cell lines. Experimental results were supported by a docking study.

Haloquinone Chloroimides as Toxic Disinfection Byproducts Identified in Drinking Water

Shuo Xu et al.

Summary: An analytical method was developed to detect highly toxic haloquinone chloroimides (HQCs) in drinking water, revealing the presence of five new disinfection byproducts (DBPs) in samples from nine water treatment plants and distribution systems. The cytotoxicity of the identified HQCs was found to be higher than or comparable to an emerging DBP, 2,6-dichloro-1,4-benzoquinone (2,6-DCBQ). This study provides the first set of occurrence and cytotoxicity data for HQC DBPs in drinking water.

Occurrence and toxicity of halobenzoquinones as drinking water disinfection byproducts

Huan Wu et al.

Summary: This study investigated the distribution and formation of halobenzoquinones in drinking water in China, comparing their cytotoxicity and genotoxicity, and analyzing the correlation of their toxicity with physicochemical parameters. It was found that the concentration of HBQs was influenced by water treatment methods and water quality parameters, with certain factors being negatively correlated with the cytotoxicity of the compounds.

A New Group of Heterocyclic Nitrogenous Disinfection Byproducts (DBPs) in Drinking Water: Role of Extraction pH in Unknown DBP Exploration

Haiyang Tang et al.

Summary: Adjusting pH prior to extraction is crucial in water sample pretreatment for studying new disinfection byproducts. This study successfully detected a new group of nitrogenous DBPs in drinking water, with recovery rates of over 50% achieved at a pH of 3.0. The optimized method allowed for accurate detection of these DBPs with limits ranging from 0.04-1.58 ng/L, and toxicity assays indicated lower toxicity compared to halogenated benzoquinones but higher than commonly known DBPs.
