Potential role of BAY11-7082, a NF-κB blocker inhibiting experimental autoimmune encephalomyelitis in C57BL/6J mice via declining NLRP3 inflammasomes

Yue Lang et al.

Summary: This study aimed to investigate the roles of NF-κB and NLRP3 in the development of MS and EAE. The results showed that BAY11-7082 could prevent and treat EAE, and the activation of NLRP3 might be related to the NF-κB pathway in EAE.

EGCG Restricts PRRSV Proliferation by Disturbing Lipid Metabolism

Peng-Wei Yu et al.

Summary: In this study, the researchers uncovered a previously unknown mechanism of how epigallocatechin gallate (EGCG) inhibits the proliferation of porcine reproductive and respiratory syndrome virus (PRRSV). They found that EGCG disrupts lipid metabolism, which leads to decreased replication of PRRSV. Additionally, EGCG also inhibits the formation of lipid droplets and the expression of key enzymes in lipid synthesis induced by PRRSV, and attenuates PRRSV-induced autophagy.

Metformin Corrects Glucose Metabolism Reprogramming and NLRP3 Inflammasome-Induced Pyroptosis via Inhibiting the TLR4/NF-κB/PFKFB3 Signaling in Trophoblasts: Implication for a Potential Therapy of Preeclampsia

Yang Zhang et al.

Summary: The study reveals the close relationship between the pathogenesis of preeclampsia and NLRP3 inflammasome-mediated pyroptosis, linked with TLR4/NF-κB signaling and PFKFB3 glycometabolism reprogramming. MET alleviates NLRP3 inflammasome-induced pyroptosis by inhibiting TLR4/NF-κB/PFKFB3 pathway.

Small molecule screening identified cepharanthine as an inhibitor of porcine reproductive and respiratory syndrome virus infection in vitro by suppressing integrins/ILK/RACK1/PKCα/NF-κB signalling axis

Chao Yang et al.

Summary: The study identified cepharanthine as a potent inhibitor of PRRSV infection by suppressing the expression of multiple proteins and the NF-?B signaling pathway. This inhibition effectively reduced infection levels and protected cells from PRRSV invasion, suggesting CEP as a promising drug for PRRS control in veterinary clinics.

NF-κB: At the Borders of Autoimmunity and Inflammation

Laura Barnabei et al.

Summary: NF-kappa B transcription factor plays a crucial role in regulating various aspects of innate and adaptive immune functions, serving as a key mediator of immune homeostasis. It is involved in central and peripheral tolerance, as well as the inactivation and destruction of autoreactive immune cells, contributing to immune tolerance. Furthermore, NF-kappa B plays a role in the pathogenesis of autoimmune diseases and has informed new therapeutic approaches for treating these conditions.
