Critical role of PAFR/YAP1 positive feedback loop in cardiac fibrosis

Tian-yu Li et al.

Summary: Abnormal activation of cardiac fibroblasts is the main cause of cardiac fibrosis. This study reveals the important roles of platelet-activating factor (PAF) and the Hippo pathway in cardiac fibrosis. PAF induces fibrosis through activation of the effector YAP1 and promotes its nuclear translocation via PAFR interaction. Furthermore, YAP1 promotes the expression of PAFR. Inhibiting PAFR or YAP1 can impede cardiac fibrosis and improve cardiac function. These findings suggest the potential of PAFR/YAP1 as therapeutic targets for cardiac fibrosis.

Circular RNAs in physiology and non-immunological diseases

Liang Chen et al.

Summary: Circular RNAs (circRNAs) are covalently closed single-stranded RNAs with unique features in their biogenesis, degradation, and transport. They have diverse molecular functions, including sponging miRNAs, regulating transcription, modulating RNA-binding proteins, and even encoding proteins. Dysregulation of circRNAs is associated with various human disorders. Further research can help resolve debates and potentially lead to biomedical applications of circRNAs.

Circular RNA circHERC4 as a novel oncogenic driver to promote tumor metastasis via the miR-556-5p/CTBP2/E-cadherin axis in colorectal cancer

Jiehua He et al.

Summary: This study found that circHERC4 was significantly upregulated in colorectal cancer tissues, associated with lymph node metastasis and advanced tumor grade, and correlated with worse survival. Silencing circHERC4 inhibited proliferation and migration of aggressive CRC cell lines and reduced metastasis in vivo. Mechanistically, circHERC4 inactivated the tumor suppressor miR-556-5p, leading to activation of the CTBP2/E-cadherin pathway promoting tumor metastasis in CRC.

The circular RNA circDLG1 promotes gastric cancer progression and anti-PD-1 resistance through the regulation of CXCL12 by sponging miR-141-3p

Dong-Liang Chen et al.

Summary: The study highlights the upregulation of circDLG1 in distant metastatic lesions and gastric cancer tissues resistant to anti-PD-1 therapy, correlating with aggressive tumor phenotype and poor prognosis. Ectopic circDLG1 expression promotes proliferation, migration, invasion, and immune evasion of gastric cancer cells. Mechanistically, circDLG1 interacts with miR-141-3p as a miRNA sponge to increase CXCL12 expression, promoting gastric cancer progression and resistance to anti-PD-1 therapy.

CircHelz activates NLRP3 inflammasome to promote myocardial injury by sponging miR-133a-3p in mouse ischemic heart

Yu Bian et al.

Summary: The study revealed that circHelz, a circRNA transcribed from the Helz gene, is significantly upregulated in ischemic myocardium and cardiomyocytes exposed to hypoxia. The overexpression of circHelz triggers NLRP3 inflammasome activation and pyroptosis, while silencing circHelz reduces these effects. Additionally, circHelz knockdown attenuates inflammasome expression, reduces myocardial injury and improves cardiac function after MI. Overexpression of miR-133a-3p prevents pyroptosis by targeting NLRP3, and circHelz functions as a sponge for miR-133a-3p. Ultimately, interfering with the circHelz/miR133a-3p/NLRP3 axis may be a promising therapeutic strategy for ischemic cardiac diseases.

Cardiac fibrosis

Nikolaos G. Frangogiannis

Summary: Myocardial fibrosis, characterized by the expansion of cardiac interstitium through deposition of extracellular matrix proteins, is a common pathophysiologic feature in various myocardial conditions. Activated fibroblasts and myofibroblasts play a central role in cardiac fibrosis, producing matrix proteins and triggering fibrogenic signalling cascades in response to stress. Immune cells, vascular cells, and cardiomyocytes can also contribute to fibrosis, while fibrotic changes may disrupt cardiac function and play a role in arrhythmogenesis.
