Role of COX-2/PGE2 signaling pathway in the apoptosis of rat ovarian granulosa cells induced by MEHP

Summary: The objective of this study was to investigate the effects of the COX-2/PGE2 pathway on MEHP-induced cell apoptosis in ovarian granulosa cells. The results showed that MEHP significantly decreased cell viability and increased apoptosis levels. PGE2 levels were markedly reduced and the expression levels of related genes were altered. Over-expression of COX-2 alleviated apoptosis and slightly increased PGE2 levels. This indicates that MEHP causes cell apoptosis in rat ovarian granulosa cells by down-regulating ovulation-related gene levels via the COX-2/PGE2 pathway.

DRIP is a risk factor for various adverse CV outcomes such as cardiac developmental toxicity, injury and apoptosis, arrhythmogenesis, metabolic disorders, vascular damage, atherosclerosis, coronary heart disease and hypertension. The underlying mechanisms involve DNA methylation, PPAR-related pathways, oxidative stress and inflammation, and disturbance of Ca2+ homeostasis. Potential preventive and therapeutic measures include the use of P-glycoprotein, heatshock proteins, antioxidants, curcumin, apigenin, beta-thujaplicin, glucagon-like peptide-1 receptor agonists, and Ang-converting enzyme inhibitors.
