Target lines for recombinase-mediated gene stacking in soybean

Summary: This study obtained five soybean target lines with suitable recombinase sites for site-specific gene stacking and successfully tested the integration of new genes into these lines. Adding new DNA to existing transgene loci can facilitate the introgression of new traits into field cultivars. The findings of this study have important implications for genetic improvement and breeding of soybean.

Target Lines for in Planta Gene Stacking in Japonica Rice

Summary: Target sites were generated in the Japonica rice genome using the Mycobacteriophage Bxb1 site-specific integration system. Single copy lines with target sites that were not close to a centromere, known genes or repetitive DNA, and were able to express the reporter gene were successfully produced. New traits were stacked into Japonica rice through gene transfer and site-specific integration.

Recombinase-mediated gene stacking in cotton

Summary: In this study, target lines suitable for gene stacking in cotton were used to stack three genes for resistance to verticillium wilt. Nine site-specific integration events were obtained from 95 transformed embryogenic calluses, and precise integration was confirmed through Southern hybridization and DNA sequence analysis. One of the regenerated plants showed resistance to verticillium wilt. These cotton target lines can facilitate precise DNA integration and introgression to field cultivars.
