Multicomponent Synthesis of Purines and Pyrimidines: From the Origin of Life to New Sustainable Approaches for Drug-Discovery Applications

Summary: It is widely recognized that the building blocks of life, purines and pyrimidines, were created through multicomponent reactions on early abiotic Earth. These structures evolved into biologically relevant molecules and served as templates for drug development. The synthetic community has dedicated efforts to replicate the abiotic synthesis of purines and pyrimidines, resulting in the development of various multicomponent synthesis methods. The importance of multicomponent synthesis of new heterocycles is highlighted by the shift towards green chemistry and pharmaceutical sustainability. This review provides an overview of the most important multicomponent approaches for synthesizing purine and pyrimidine derivatives for potential pharmacological applications.

N-Aminoacyl-3-amino-nido-carboranes as a Group of Boron-Containing Derivatives of Natural Amino Acids

Summary: A new group of nido-carboranyl derivatives of natural (S)-amino acids containing from 9 to 18 boron atoms was successfully synthesized by acylation of 3-amino-1,2-dicarba-closo-dodecaborane followed by deboronation. The proposed method utilizes readily available reagents and is convenient for the synthesis of enantiopure nido-carboranyl derivatives of amino acids with various side chains, including water-soluble boron-containing amino acids (17 examples).

Substituted Purines as High-Affinity Histamine H3 Receptor Ligands

Summary: In this study, a series of new purine derivatives were synthesized and screened for their affinity to H3R. Two compounds showed high affinity to H3R and were more potent than the reference drug. Molecular modeling studies revealed the importance of the purine ring in enhancing affinity through hydrogen bonding. These findings suggest that these molecules have promising potential as H3R ligands.

Carborane-Containing Folic Acid bis-Amides: Synthesis and In Vitro Evaluation of Novel Promising Agents for Boron Delivery to Tumour Cells

Summary: This work develops a new type of folic acid bis-amides containing boron, which exhibit high water solubility and low cytotoxicity. The experimental results demonstrate the effective delivery of boron to tumor cells by these compounds, highlighting their potential for application in boron neutron capture therapy.

Carborane-containing amino acids and peptides: Synthesis, properties and applications

Special attention is given to the preparation of enantiopure conjugates and the chirality origins in these compounds, aiming to stimulate further fundamental research in this field.

Summary: A series of modified nucleoside triphosphates bearing dicarba-nido-undecaborate or metal bis(dicarbollide) groups were prepared and utilized for DNA synthesis and redox labeling. The combination of these redox labels allowed for the first fully orthogonal redox coding of all four canonical nucleobases, enabling determination of nucleobase composition of short DNA stretches with electrochemical readout in one simple primer extension experiment.

Interaction of Adenosine, Modified Using Carborane Clusters, with Ovarian Cancer Cells: A New Anticancer Approach against Chemoresistance

Summary: The study focused on designing and synthesizing adenosine conjugates with metallacarboranes containing iron, cobalt, or chromium to influence the cisplatin sensitivity of ovarian cancer cells. The iron-containing metallacarborane conjugates demonstrated the best anticancer activity, sensitizing resistant cells without inducing cross-resistance to other chemotherapy drugs. This research highlights the potential of using metallacarborane-modified adenosine to enhance the sensitivity of ovarian cancer cells to cisplatin in combination therapy.
