Summary: The impact of co-mutations on the response to EGFR TKIs in different types of EGFR-mutant lung cancer remains unclear. This retrospective study analyzed the outcomes of 137 patients receiving frontline EGFR TKI therapy and found that co-mutations played a role in the shorter time to treatment failure in the L858R cohort compared to the exon 19 deletion cohort.

Summary: This is the first meta-analysis to evaluate the clinical outcomes and acquired T790M mutation frequency in relation to EGFR 19del subtypes with EGFR-TKI therapy. Patients with a deletion starting from E746 showed better overall survival compared to other subtypes, and patients with the E746_A750del subtype had a higher frequency of acquired T790M mutation.

Additionally, advancements in assay sensitivity have allowed for the detection of ctDNA at very low allele frequencies, aiding in the measurement of minimal residual disease and improving the efficiency of lung cancer screening programs.
