Ultra-Small High-Entropy Alloy Nanoparticles: Efficient Nanozyme for Enhancing Tumor Photothermal Therapy

Summary: High-entropy alloy nanoparticles (HEANPs) are attracting attention for their tunability and potential in bioapplication. However, it is challenging to prepare ultra-small high-entropy alloy nanoparticles (US-HEANPs) due to their instability. In this study, PtPdRuRhIr US-HEANPs were fabricated and used as multifunctional nanoplatforms for tumor treatment. The US-HEANPs exhibited peroxidase-like activity and photothermal conversion effect, which effectively ablated cancer cells and treated tumors. This work provides new insights into HEANPs fabrication and opens up research on high-entropy nanozymes and their biomedical applications.

Summary: In this study, a metal-ligand cross-linking strategy was developed for the fabrication of ferric ion-dopamine coordination hierarchical superstructures. Various superstructures with complex morphologies were synthesized, and the formation mechanism was explored. The hierarchical superstructure was used as peroxidase-like ligase mimics to enhance tumor photothermal treatment, and it showed effective tumor cell ablation in vitro and in vivo. This work opens new horizons in hierarchical superstructures and has great potential in nanozymology, biomedical science, and catalysis.

Summary: The development of high-efficiency nanozymes with excellent oxidase-like and peroxidase-like enzyme activities is significant for enhancing the oxidation of ascorbate to improve cancer cell ablation efficacy. The fabricated nanozyme can rapidly and completely oxidize ascorbate within 20 minutes, leading to tumor cell death through the generation of superoxide free radicals and hydrogen peroxide.Overall, the research shows a promising enhancement in anti-tumor efficacy through ascorbate oxidation catalyzed by the developed nanozyme.
