Mitochondrial Membrane Potential Independent Near-Infrared Mitochondrial Viscosity Probes for Real-Time Tracking Mitophagy

Summary: Mitophagy is a crucial cellular process that plays vital roles in regulating cellular metabolism and mitochondrial quality control. Mitochondrial viscosity, closely associated with mitochondrial status, is a key microenvironmental index. To monitor mitophagy and mitochondrial viscosity changes, a probe called Mito-3 was developed. Mito-3, along with other probes, showed sensitivity to viscosity changes and fluorescence enhancement. These probes were successfully used to locate and visualize mitochondria, as well as monitor mitochondrial viscosity changes in cells. Additionally, Mito-3 was applied to visualize the mitophagy process induced by starvation and revealed an increase in mitochondrial viscosity during mitophagy.

An ultra-photostable and leakage-free fluorescent probe for long-term and high-fidelity mitochondria visualization and tracking

Summary: In this study, we report a cationic probe DQPI-12 with a long alkyl chain that can effectively target mitochondria and has excellent leakage-free performance. DQPI-12 also exhibits superior photostability compared to commercial mitochondria probes and those reported by others. The inherent characteristics of DQPI-12 enable long-term and high-fidelity visualization of mitochondria in a special microscopic field, allowing real-time and in-situ tracking of mitochondrial dynamics and mitophagy.
