Y-shaped walker with abundant recognition domians mediated ultrasensitive electrochemical biosensor for miRNA-21 detection

Summary: In this study, an ultrasensitive electrochemical biosensor was constructed using a high-efficiency Y-shaped DNA walker with abundant recognition domains. The Y-shaped DNA walker could be synthesized through rolling circle amplification with the assistance of padlock, and it showed significantly higher cleavage efficiency compared to conventional Y-shaped walkers. The biosensor achieved ultrasensitive detection of miRNA-21 with a detection limit of 10.6 aM, indicating its potential for sensitive detection of biomarkers and early diagnosis of diseases.

The AND-gated ratio fluorescence of r-AgNC over g-AgNC can only be lit up when both To and Tr are overexpressed in SARS-CoV-2, enabling highly specific-inputs binding and sensitive dose-dependence.
