A mesothelium divides the subarachnoid space into functional compartments

Kjeld Mollgard et al.

Summary: The central nervous system is lined by four meningeal layers, including a newly discovered layer called subarachnoid lymphatic-like membrane (SLYM), which resembles mesothelial membrane found in peripheral organs. SLYM encloses blood vessels and contains immune cells. It allows direct exchange of small solutes between cerebrospinal fluid and venous blood, similar to arachnoid granulations in mice. The functional characterization of SLYM provides important insights into brain immune barriers and fluid transport.

The genetic etiology of hearing loss in Japan revealed by the social health insurance-based genetic testing of 10K patients

Shin-ichi Usami et al.

Summary: This study analyzed 10,047 samples from social health insurance-based genetic testing using the same platform and filtering algorithm. It clarified the genetic epidemiology of hearing loss from congenital/early-onset deafness to late-onset hearing loss, revealing different diagnostic rates and causative genes for each age group.

Genome-wide association meta-analysis identifies 48 risk variants and highlights the role of the stria vascularis in hearing loss

Natalia Trpchevska et al.

Summary: Hearing loss is a significant contributor to disability and a risk factor for dementia. By analyzing genetic data and mapping it to specific cells in the cochlea, this study identifies novel loci and highlights the importance of the stria vascularis in hearing impairment, offering potential targets for therapeutic intervention.

The glymphatic system: implications for drugs for central nervous system diseases

Terhi J. Lohela et al.

Summary: Evidence for a fluid clearance pathway called the glymphatic system in the central nervous system has increased in the past decade. The glymphatic system clears metabolic waste from the brain interstitium by directing the flow of cerebrospinal fluid. It can be modulated by drugs such as anaesthetics and non-pharmacological factors like sleep, and its dysfunction is associated with central nervous system disorders like Alzheimer's disease. While primarily studied in rodents, multiple neuroimaging modalities suggest the existence of a similar transport system in the human brain. This review provides an overview of the evidence for the glymphatic system, its role in disease, and opportunities for therapeutic intervention.

Axon-glia interactions in the ascending auditory system

David C. Kohrman et al.

Summary: This review focuses on the roles of glial cells and glia-like cells in the ascending auditory pathway and their impact on the development, maintenance, and modulation of neural circuits and transmission in the auditory system. The molecular mechanisms of these interactions and their effects on auditory dysfunction associated with pathologies of each cell type are also discussed.

Advances and challenges in adeno-associated viral inner-ear gene therapy for sensorineural hearing loss

Kamakshi Bankoti et al.

Summary: There is a growing focus on treating the root cause of sensorineural hearing loss instead of just managing secondary characteristic features. Gene delivery has emerged as a promising strategy for the biological treatment of hearing loss associated with genetic dysfunction, with successful proof-of-principle examples in animal models but further progress needed for clinical application in humans.
