Mitochondria ROS and mitophagy in acute kidney injury

Summary: Mitophagy is a crucial mitochondrial quality control mechanism that removes damaged mitochondria and reactive oxygen species (ROS) production. The relationship between mitochondrial oxidative stress, ROS production, and mitophagy is closely intertwined and involved in various pathological conditions of acute kidney injury (AKI). The complex process of mitophagy in mammals involves multiple pathways and its interaction with different types of cell death, such as apoptosis, pyroptosis, and ferroptosis in kidney injury, remains unclear. This review discusses recent advances in understanding the relationship between ROS and mitophagy, the different pathways of mitophagy, the interplay between mitophagy and cell death, and their significance in the pathogenesis of AKI.

The potential ecological risk of veterinary pharmaceuticals from swine wastewater on freshwater aquatic environment

Summary: The transport of pharmaceutical residues in the aquatic ecosystem has negative environmental and public health impacts due to the bioactivity of trace antibiotics. This study investigated the presence of three veterinary pharmaceuticals in piggery effluent and assessed the ecological risk on freshwater organisms. The results showed that high doses of piggery effluent and mixtures of veterinary pharmaceuticals pose a high risk to freshwater ecosystems.

Cutaneous Nrf2-Keap1 pathway modulation by environmental factors: The Dead Sea area as a test case

Summary: The skin can benefit from exposure to moderate environmental stressors, such as those found in the Dead Sea area, which activates the Nrf2-Keap1 pathway and protects against oxidative damage. The unique combinations of moderate stressors in the Dead Sea, including high-water salinity, high atmospheric pressure, and unique solar radiation, generate moderate oxidative stress in the skin, leading to the induction of intracellular electrophiles and the activation of the protecting Nrf2-Keap1 pathway. This activation results in the induction of phase II enzymes and lower apoptotic activity compared to nearby locations at higher altitudes.

PINK1-Mediated Mitophagy Promotes Oxidative Phosphorylation and Redox Homeostasis to Induce

Summary: This study demonstrates that lung adenocarcinoma cells in drug-tolerant state undergo metabolic switch from glycolysis to oxidative phosphorylation. PTEN-induced kinase 1 (PINK1) upregulation mediates mitophagy to maintain mitochondrial homeostasis and promote survival of drug-tolerant cells, leading to poor prognosis. Inhibition of MAPK pathway results in MYC-dependent transcriptional upregulation of PINK1 and activation of mitophagy. Mitophagy inhibition using clinically applicable chloroquine or depletion of PINK1 can eradicate drug tolerance and achieve complete response to MAPK inhibitors.

Dysregulation of mitochondrial dynamics, mitophagy and apoptosis in major depressive disorder: Does inflammation play a role?

Summary: Recent studies have shown that mitochondrial dysfunction and dysregulated neuroinflammatory pathways play a role in major depressive disorder (MDD). This study found differences in markers of mitochondrial dynamics, mitophagy, autophagy, and apoptosis in MDD patients compared to healthy controls, with higher levels of specific proteins associated with mitochondrial dysfunction in MDD patients. Furthermore, inflammation and protein levels related to mitochondrial dynamics and mitophagy were found to influence the severity of depressive symptoms in MDD patients.

Veterinary antibiotics can reduce crop yields by modifying soil bacterial community and earthworm population in agro-ecosystems

Summary: Veterinary antibiotics are widely used in animal farming and their presence in agroecosystems can have negative effects on soil bacteria, earthworms, and soil nutrients. This study found that veterinary antibiotics inhibit earthworm abundance, decrease bacterial diversity, and reduce soil nutrient availability. High-dose exposure to antibiotics also increases the abundance of plant pathogenic bacteria. The effects of antibiotics on crop yields are mediated by changes in soil microbes and earthworms, but the direct toxicity effects of antibiotics on crop yields are stronger. The tradeoffs between antibiotics and agroecosystem functions are dose-dependent.
