A Cu+/Thiourea Dendrimer Achieves Excellent Cytosolic Protein Delivery via Enhanced Cell Uptake and Endosome Escape

Summary: This study proposes a Cu+ -based coordination polymer for highly efficient and robust delivery of intracellular proteins. The polymer efficiently binds cargo proteins through a combination of coordination, ionic and hydrophobic interactions. The incorporation of Cu+ ions greatly improves cellular uptake and endosomal escape, enabling successful delivery of various structurally diverse proteins into different cell lines with reserved bioactivities. This study provides a new type of coordination polymers for cytosolic delivery of biomacromolecules.
