The results demonstrate how ligand-encoded chiral information is transferred to the alpha-/beta-sites of the resulting alpha-AAs and how the mode of regio-/stereoselection is altered by differences in transition-metal-stabilized coupling partners.

Summary: An enantioselective (3+2) cycloaddition reaction has been reported for the synthesis of chiral polysubstituted pyrrolidines. The reaction displays opposite regioselectivity with excellent enantioselectivities and yields. Interestingly, changing the alpha-substituents of the iminoesters results in the formation of normal (3+2) cycloaddition products with excellent enantioselectivities as well.

Catalytic enantioselective intramolecular 1,3-dipolar cycloaddition of azomethine ylides with fluorinated dipolarophiles

Summary: An enantioselective synthesis of polycyclic fluorinated pyrrolidines has been achieved by Cu-catalyzed intramolecular 1,3-dipolar cycloaddition. The method enables the formation of desired products with high yields and up to 99% enantiomeric excess, indicating the excellent activating role of fluoroalkyl substituents in this transformation.

Combined experimental and computational mechanistic studies supported a Co-II catalytic cycle involving migratory insertion and sigma-bond metathesis processes, with enantioselectivity possibly originating from the steric effect between the phenyl groups of the ligand and the substrate.

Summary: Experimental and theoretical studies have investigated the 1,3-dipolar cycloaddition of alpha-fluoro-alpha, beta-unsaturated arylketones and azomethine ylides. The reaction proceeds in a diastereodivergent fashion, with the endo-selective pathway achieved using an Ag(I) catalyst. Theoretical DFT calculations suggest that the coordination between the silver atom and the carbonyl oxygen atom of the enone drives the endo-selectivity.

Asymmetric catalytic 1,3-dipolar cycloaddition of α-diazoesters for synthesis of 1-pyrazoline-based spirochromanones and beyond

Summary: In this study, a highly enantioselective 1,3-dipolar cycloaddition of alpha-substituted diazoesters with exocyclic enones was achieved using a chiral scandium(III)/N,N'-dioxide complex as the catalyst. The protocol provided an efficient route to optically active products with good outcomes, including up to 97% yield and 98% ee with >95:5 dr. Additionally, enantioenriched 2-pyrazoline-based spirochromanones were accessible by switching alpha-substituted diazoesters to alpha-diazoacetates. Further specific transformations of chiral pyrazoline-based spiro-compounds to spirocyclopropane derivatives were also disclosed in the study.
