The authors proposed an alternative mechanism of insertion, reconciling the ability of type III transmembrane proteins to bypass an ipomoeassin-F-mediated blockade of membrane integration.

Architecture of the active post-translational Sec translocon

Summary: The article presents a cryo-EM structure of the yeast Sec complex bound to a substrate and a crystal structure of the Sec62 cytosolic domain. The interaction between Sec62 domain and Sec63 C-terminus was also mapped.

Summary: Liquid-like condensates in cells are not necessarily sphere-like, as various condensates with filamentous morphology have been observed. One example is the TIS granule network, which shares a large surface area with the rough endoplasmic reticulum and plays a crucial role in membrane protein trafficking. Reconstitution experiments have shown that a multivalent RNA-binding protein and RNAs with large unstructured regions can form a pervasive intermolecular interaction network that acts as a condensate skeleton.

Specific subsets of cytosolic and ER-targeted mRNAs can be recruited into SGs, which form in close proximity to or directly associated with the ER membrane. ER-targeted and cytosolic mRNAs can be recruited into ER-associated SGs, with a functional link between gene transcriptional state and SG biogenesis.

Alternative 3′ UTRs play a widespread role in translation-independent mRNA association with the endoplasmic reticulum

Summary: Transcripts encoding membrane and secreted proteins can associate with the ER through translation-dependent or translation-independent mechanisms. Certain functional groups of genes, such as those involved in cell signaling, tend to have higher TiERA potentials, indicating the importance of ER association for their mRNA metabolism. Changes in transcript features, such as alternative polyadenylation isoforms and 3' UTR lengthening during cell differentiation, can affect the TiERA potential of transcripts. Competing with translation-dependent ER association, TiERA suggests a limited space on the ER for mRNA association.
