Anti-protein arginine deiminase antibodies are distinctly associated with joint and lung involvement in rheumatoid arthritis

Summary: This study aimed to assess clinical features, ACPA, and anti-PAD antibodies in RA patients with articular involvement and ILD. The results showed that anti-PAD4 antibodies were significantly associated with radiographic injury and erosions, while ACPA levels were associated with erosive disease. Additionally, patients with both anti-PAD3 and anti-PAD4 antibodies had more joint erosions than those with only anti-PAD4 antibodies or negative for both.

Neutrophil extracellular traps in systemic autoimmune and autoinflammatory diseases

Summary: Systemic autoimmune diseases occur when the immune system fails to recognize self from non-self, resulting in significant morbidity and mortality. Neutrophils, especially the neutrophil extracellular traps they release, play a crucial role in the initiation and perpetuation of these diseases, leading to organ damage. Dysregulation of neutrophil cell death can alter autoantigens and trigger complex inflammatory responses. Studies have identified specific neutrophil subsets associated with systemic autoimmune disorders that contribute to vascular damage and oxidative stress. Advances in technology have enhanced our understanding of neutrophil biology and dysregulation. This review provides an overview of the roles of neutrophils in systemic autoimmune and autoinflammatory diseases and explores potential therapeutic targets.

Summary: Based on the epidemiological link between periodontitis and rheumatoid arthritis (RA), this study investigated the production of anti-citrullinated protein antibodies (ACPA) triggered by the periodontal bacterium Porphyromonas gingivalis in the gum mucosa of early RA patients. The findings suggest that immunity to P. gingivalis derived citrullinated antigens in the inflamed gum mucosa may contribute to the presence of ACPA in RA patients through mechanisms of molecular mimicry.
