Reproductive and developmental toxicity assessments of LPM3480392, a novel mu opioid receptor biased agonist in rats

Synthesis, biological, and structural explorations of a series of μ-opioid receptor (MOR) agonists with high G protein signaling bias

Summary: Biased agonism refers to compounds' ability to preferentially activate certain signaling pathways and avoid adverse signaling pathways in a ligand-dependent manner for some G-protein-coupled receptors. In this study, a series of derivatives with potent highly biased MOR agonism were rationally designed and synthesized through modification and structure-activity relationship study of TRV130. The novel molecule LPM3480392 demonstrated improved in vitro biased agonism, good brain penetration, and produced potent antinociceptive effect with reduced respiratory suppression compared to TRV130.

Enduring consequences of perinatal fentanyl exposure in mice

Summary: This study found that perinatal exposure to fentanyl in mice resulted in withdrawal behavior and behavioral abnormalities in early adulthood, similar to humans born with NOWS. These effects were seen in terms of changes in behavior and physiology that normalized as the mice reached adulthood.

Summary: Studies on the long-term exposure of hydroxyurea in pregnant Sprague Dawley dams and their offspring revealed that high doses of hydroxyurea had negative effects on offspring development and body weight, but had minimal maternal toxicity. Additionally, gestational transfer of hydroxyurea was observed, while evidence of lactational transfer was minimal.

Effects of maternal consumption of morphine on rat skeletal system development

Summary: The study showed that maternal consumption of morphine has detrimental effects on skeletal development in rat offspring, with more pronounced effects on seven-day-old infants. Morphine reduced the thickness and cell number of growth plates, as well as the expression of key genes involved in bone formation. These findings suggest that morphine dependence during pregnancy may impair bone development in post-natal offspring.
