Defects in placental syncytiotrophoblast cells are a common cause of developmental heart disease

Summary: This study demonstrates that the placenta might be a significant cause of congenital heart defects, using mouse genetic tools.

The Trophoblast Compartment Helps Maintain Embryonic Pluripotency and Delays Differentiation towards Cardiomyocytes

Xiang Zhao et al.

Summary: Normal developmental progression relies on close interactions between the embryonic and extraembryonic lineages in the pre- and peri-gastrulation stage conceptus. Trophoblast cells of the extraembryonic ectoderm play a role in delaying cardiomyocyte differentiation dynamics and prolonging pluripotency in embryonic stem cells (ESCs). These findings provide insights into the intricate crosstalk between different compartments of the early conceptus and have implications for synthetic embryo research.

Trophoblast inclusions and adverse birth outcomes

Morgan R. Firestein et al.

Summary: Objective trophoblast inclusions are associated with adverse birth outcomes and specific placental pathologies. The presence of trophoblast inclusions is significantly higher in preterm and stillborn deliveries compared to full-term liveborn deliveries. These findings suggest the need for further investigation into the complex mechanisms underlying trophoblast inclusions.

Prediction of stillbirth: an umbrella review of evaluation of prognostic variables

R. Townsend et al.

Summary: This study collated and evaluated factors associated with stillbirth to identify variables relevant to prediction model development. The most convincing associations were found with age, parity, and prior adverse pregnancy outcomes. Tests like PAPP-A, PlGF, and Uterine artery Doppler showed good performance in predicting stillbirth.

Stillbirth and fetal anomalies: secondary analysis of a case-control study

S. L. Son et al.

Summary: The study found that 23.4% of stillbirths had one or more major anomalies, compared to 4.3% of live births. The presence of anomalies increased the odds of stillbirth, with a higher association observed with an increasing number of anomalies. Regardless of the affected organ system, the presence of an anomaly was found to increase the odds of stillbirth.

Exome sequencing analysis on products of conception: a cohort study to evaluate clinical utility and genetic etiology for pregnancy loss

Chen Zhao et al.

Summary: Exome sequencing showed clinical utility in identifying monogenic causes of pregnancy loss, detecting pathogenic variants and providing insights for genetic counseling and management of subsequent pregnancies. Detection rate increased by including variants of uncertain significance, with discovery of novel variants helping to understand molecular mechanisms leading to fetal death.

Summary: The study identified four different subtypes of trophoblast inclusions (TIs) in placentas, including inclusionoids, inclusions, calcified inclusions, and calcified bodies. The number of TIs present and the timing of their expression in placentas may be related to clinical outcomes.

Can information regarding the index stillbirth determine risk of adverse outcome in a subsequent pregnancy? Findings from a single-center cohort study

Nicole Graham et al.

Summary: Placental examination at the time of stillbirth is crucial for predicting the risk of adverse outcomes in subsequent pregnancies, while information about maternal characteristics and classification of the cause of stillbirth does not provide significant prognostic information.

Summary: The study found that monozygotic twins showed significantly less variation in the average number of TIs per slide compared to dizygotic twins, suggesting that TIs are intrinsic to the genetics of the fetus rather than the uterine environment.
