Quiescent human glioblastoma cancer stem cells drive tumor initiation, expansion, and recurrence following chemotherapy

Xuanhua P. Xie et al.

Summary: This study confirms the existence of quiescent cancer stem cells in glioblastoma that can evade anti-proliferative therapies. By analyzing the functional characteristics of glioblastomas from genetically engineered mice, essential quiescent stem-like cells that can be directly isolated from tumors were identified. A specific gene expression signature related to quiescent cancer stem cells was found in both primary and recurrent human glioblastomas. These findings provide insights into the failure of conventional treatment and lay the foundation for alternative therapies.

Integrating computational pathology and proteomics to address tumor heterogeneity

Anglin Dent et al.

Summary: Despite advances in cancer biology, precision medicine trials face challenges due to molecular inconsistencies and heterogeneous tumor biology. Integrating mass-spectrometry-based global proteomics and computational imaging can overcome these challenges of biologic variation in cancer.

Interactions between cancer cells and immune cells drive transitions to mesenchymal-like states in glioblastoma

Toshiro Hara et al.

Summary: The mesenchymal subtype of glioblastoma is believed to be influenced by both cancer cell-intrinsic alterations and external cellular interactions, with macrophages inducing a transition of glioblastoma cells into mesenchymal-like states. This transition is mediated by macrophage-derived oncostatin M (OSM) interacting with its receptors on glioblastoma cells, leading to the activation of STAT3. This study also reveals that mesenchymal-like glioblastoma states are associated with increased expression of a mesenchymal program in macrophages and enhanced cytotoxicity of T cells, suggesting potential therapeutic implications.
