L1CAM immunocapture generates a unique extracellular vesicle population with a reproducible miRNA fingerprint

Rachael Anne Dunlop et al.

Summary: MicroRNAs(miRNAs) have potential as diagnostic and prognostic biomarkers, but inconsistencies across studies have hindered their clinical application. Differences in miRNAs associated with specific diseases and regulation direction reflect the heterogeneity of pathologies or different protocols. This study aimed to determine if a unique extracellular vesicle fraction could improve the reproducibility of miRNA fingerprint diagnostic of ALS.

Review on Strategies and Technologies for Exosome Isolation and Purification

Jiaci Chen et al.

Summary: Exosomes, as nano-sized extracellular vesicles, play an important role in cell-specific material transfer and have potential applications in prognosis, diagnosis, and treatment. Existing exosomal separation techniques have limitations in terms of complexity, time consumption, and low purity, highlighting the need for efficient and simple isolation methods. Microfluidic chips have emerged as a promising technology for exosome isolation. A comprehensive review covering exosomal properties, conventional technologies, emerging isolation strategies, and large-scale production of engineered exosomes is essential.

Selective immunocapture reveals neoplastic human mast cells secrete distinct microvesicle- and exosome-like populations of KIT-containing extracellular vesicles

Annika Pfeiffer et al.

Summary: This study isolated and characterized subpopulations of KIT-containing extracellular vesicles (EVs) released by neoplastic human mast cells (huMCs). The KIT-EVs were found to have distinct protein profiles and were involved in signaling, immune responses, and cell migration. The study also revealed that KIT-EVs are transferred via heterogeneous microvesicle- and exosome-like EVs. The selective immunocapture of KIT-EVs will facilitate the enrichment of specific huMC-derived EVs from complex human samples and provide insights into their in vivo functions and potential as biomarkers.

Marie-Nicole Theodoraki et al.

Summary: Exosomes, also known as small extracellular vesicles or sEV, are a system of intercellular communication. Quantification of exosome cargo using on-bead flow cytometry is essential for understanding their role in information transfer and human disease. This method is standardized for protein detection and quantitation in exosomes isolated from cell supernatants or plasma of cancer patients, providing valuable insights into the surface and intravesicular content of these vesicles.
