Computational design of constitutively active cGAS

Summary: Cyclic GMP-AMP synthase (cGAS) is an important pattern recognition receptor involved in the innate immune response. By binding to dsDNA, cGAS is activated to produce cGAMP, which triggers a strong interferon response. Researchers have successfully designed constitutively active cGAS variants that can function independently of dsDNA. These variants showed therapeutic activity in tumor regression when expressed in tumor cells. This study provides insights into enzyme stabilization and offers potential applications in understanding inflammatory diseases.

Computational protein design repurposed to explore enzyme vitality and help predict antibiotic resistance

Summary: Physics and physical chemistry play an important role in computational protein design, offering molecular mechanics scoring functions, thorough sampling methods for equilibrium ensembles, and different levels of approximation for conformational flexibility. Recent advancements have successfully used physics-based folded state energy to redesign a small protein. Adaptive Monte Carlo or molecular dynamics schemes have been discovered to quantitatively populate protein variants based on their ligand-binding free energy or catalytic efficiency. Molecular dynamics has been employed to study backbone flexibility. Additionally, improvements have been made in implicit solvent models, the application of polarizable force fields, and the acquisition of physical insights.
