A review on pectinase properties, application in juice clarification, and membranes as immobilization support

Summary: Pectinase enzymes are biological compounds that degrade pectic compounds in fruit juices, making them excellent biocatalysts for juice clarification. However, the poor stability and nonreusability of pectinases limit their effectiveness. Immobilization techniques, particularly using membranes, have been shown to enhance the performance of enzymes. This review highlights the importance of physicochemical properties and kinematic properties of pectinases in relation to juice processing parameters and discusses the potential of membrane immobilization as a support for pectinase.

High-yield production of acidic pectin lyase PNLZJ5B for juice processing

Summary: A pectin lyase gene pnlzj5b from Aspergillus niger ZJ5 was successfully identified and overexpressed in Pichia pastoris. The recombinant protein PNLZJ5B showed high activity towards citrus pectin and its expression efficiency was improved through codon optimization. PNLZJ5B exhibited optimal activity and stability at specific temperature and pH, and it also showed positive response to the degree of pectin methylation and had a higher juice yield compared to commercial pectin lyase. These favorable enzymatic properties make PNLZJ5B potentially useful in juice processing and other food-related applications.

Genome mining of Fusarium reveals structural and functional diversity of pectin lyases: a bioinformatics approach

Summary: In this study, 52 pectin lyase genes were predicted and characterized in six species of Fusarium. The protein sequences showed variability in terms of amino acid residues, molecular weight, and pI. Multiple sequence alignment and phylogenetic analysis revealed conserved residues and distinct groups. Three-dimensional structure prediction and analysis showed diverse structural motifs. The study provides important insights into the diversity and potential applications of pectin lyase enzymes.

A Health-Friendly Strategy for Covalent-Bonded Immobilization of Pectinase on the Functionalized Glass Beads

The polyaldehyde pullulan showed better immobilization efficiency and stability compared to glutaraldehyde, while both systems exhibited similar temperature stability. The results suggest that polyaldehyde pullulan could be a promising alternative to glutaraldehyde for enzyme immobilization.

Summary: Pectate lyases and pectin lyases play essential roles in various biotechnological applications, such as textile industry, paper making, juice clarification. They can effectively cleave the alpha-1,4-glycosidic bond of pectin molecules to produce pectin oligosaccharides, with advantages of good enzymatic selectivity, less by-products, mild reaction conditions and high efficiency. However, there is currently no comprehensive review summarizing recent advances in this field.

Partial Purification and Characterisation of Pectinase Produced by Aspergillus niger LFP-1 Grown on Pomelo Peels as a Substrate

Summary: In this study, pectinase was successfully produced by Aspergillus niger LFP-1 on pomelo peels as a sole carbon source. The enzyme was purified using ammonium sulfate precipitation, anion-exchange column chromatography, and gel filtration column chromatography, with a final yield of 0.01% and a molecular weight of 48 kDa. The purified pectinase exhibited optimal pH and temperature at 3.5 and 50 degrees C, and was stable in a pH range of 3.5 to 4.5 and at high temperatures (40-50 degrees C) for 100 min. Additionally, pectin from citrus fruit and Co2+ were identified as the best substrate and inducer to enhance pectinase yield, respectively.
