Structure determination of high-energy states in a dynamic protein ensemble

Summary: This article presents a method for high-resolution structure determination of lowly populated states by coupling NMR spectroscopy-derived pseudocontact shifts (PCSs) with Carr-Purcell-Meiboom-Gill (CPMG) relaxation dispersion (PCS-CPMG). The method accurately determines high-energy structures and defines the corresponding kinetics and thermodynamics. Application of the method to adenylate kinase (Adk) during catalysis resolves a longstanding controversy and suggests a two-step mechanism for catalysis. Unlike other methods, this approach is particularly effective for smaller systems and lowly populated states.
