Structure of the human DICER-pre-miRNA complex in a dicing state

Summary: It has been found that hDICER undergoes large conformational changes to attain the active state and recognizes pre-miRNAs through both sequence-independent and sequence-specific recognition of the newly identified 'GYM motif'. Cancer-associated mutations in the 5' pocket residues are identified, which impair miRNA biogenesis. This study reveals the mechanisms of hDICER and provides insights into hDICER-related diseases.

Drug-Like Small Molecules That Inhibit Expression of the Oncogenic MicroRNA-21

Summary: We have discovered drug-like small molecules that can bind specifically to the precursor of oncogenic and pro-inflammatory microRNA-21 with high affinity. These molecules induce structural changes in the RNA, leading to specific inhibition of miRNA processing. The most potent compound reduces cellular proliferation and miR-21 levels in cancer cell lines without inhibiting kinases or classical receptors, providing a potential avenue for therapeutic development in diseases with abnormal miR-21 expression.

Secondary structure RNA elements control the cleavage activity of DICER

Summary: By conducting experiments on more than 20,000 different shRNAs, we discovered a single-nucleotide bulge (22-bulge) that enhances the cleavage activity of DICER on shRNAs and pre-miRNAs. Additionally, polymorphism-edited 22-bulges were found to control the cleavage sites of DICER on pre-miRNAs, thereby regulating their functions.

A Slow Dynamic RNA Switch Regulates Processing of microRNA-21

Summary: This study reveals a new mechanism of RNA regulation, showing that the conformational rearrangement at the Dicer cleavage site of pre-miR-21 can regulate the processing efficiency of microRNA. By manipulating this dynamic switch, it is possible to selectively stabilize the unprocessed or processed states of pathogenic microRNAs, providing a potential chemical approach for their suppression or activation.

RNA structure probing reveals the structural basis of Dicer binding and cleavage

Summary: Sequencing methods like icSHAPE have been developed to probe RNA structures transcriptome-wide in cells. The authors of this study introduced icSHAPE-MaP, a method combining in vivo selective 2'-hydroxyl acylation and mutational profiling to probe intact RNA structures, revealing distinct structural categories of Dicer substrates in correlation to both their binding affinity and cleavage efficiency. By analyzing RNA structural data using tertiary structural modeling constrained by icSHAPE-MaP, the study found that spatial distance measuring is an influential parameter for Dicer cleavage-site selection.
