Synthesis and biological evaluation of novel [1,2,3]triazolo-pyrrolo [1,2-a]pyrido[4,3-d]pyrimidines as EGFR targeting anticancer agents

Summary: In this study, a series of fused [1,2,3]triazolo-pyrrolo[1,2-a]pyrido[4,3-d]pyrimidines were synthesized via click chemistry and carbon-carbon bond coupling. The structures of these compounds were characterized using various analysis techniques. The synthesized compounds were evaluated for their anticancer activity against different human cancer cell lines and compared with a standard drug. The most active compounds demonstrated superior potency in inhibiting the tyrosine kinase EGFR and showed promising in silico pharmacokinetic profiles.

Synthesis and Biological Evaluation of Novel N-[3-fluoro-4-(morpholin-4-yl)phenyl]thiazol-2-amine Derivatives as Potent Antibacterial and Anticancer Agents and ADMET

Summary: This report presents a series of novel N-[3-fluoro-4-(morpholin-4-yl)aryl]thiazol-2-amine derivatives (6a-6l) that were synthesized and evaluated for their antibacterial and anticancer activity. Among them, compounds 6i and 6j exhibited potent antibacterial activity against Staphylococcus aureus and Staphylococcus epidermidis, and also demonstrated inhibitory effects on biofilm growth. Compounds 6h and 6k showed high anticancer activity against the A-549 cancer cell line, with compound 6k exhibiting similar potency to the reference drug erlotinib against EGFR.

Summary: A one-step synthesis of fused Benzo[d]thiazolo-1,2,3-triazoles was developed using sulfonylazides and iodoalkyne. Several compounds showed superior activity against A-549 and good activity against MCF-7. In vitro and in silico studies demonstrated promising potency of the compounds against EGFR receptors.

Quinoline conjugated 2-azetidinone derivatives as prospective anti-breast cancer agents: In vitro antiproliferative and anti-EGFR activities, molecular docking and in-silico drug likeliness studies

Summary: Two series of quinoline conjugated 2-azetidinones were evaluated for their antiproliferative potential against breast cancer cell lines. The compounds showed higher activity against MCF7 cells than MDA-MB-231 cells and some compounds exhibited activity superior to the standard erlotinib. The most promising compounds showed inhibitory activity against EGFR tyrosine kinase and selective toxicity against the cancer cell lines.

One-Pot Synthesis of Some New Phthalazine-Piperazine-1,2,4-Oxadiazole Hybrids: Anticancer Evaluation, Molecular Docking and ADMET Studies

Summary: In this study, new phthalazine-piperazine-1,2,4-oxadiazole hybrids were synthesized and evaluated for their anti-cancer activity against three human cancer cell lines. Compound 4d showed the highest activity and strong binding to EGFR receptor. Additionally, the compounds exhibited low cytotoxicity towards normal cells and complied with pharmacokinetic rules.

Ramachary-Bressy-Wang [3+2]cycloaddition reaction: Synthesis of fully decorated 1,2,3-triazoles as potent anticancer and EGFR inhibitors

Summary: This study developed a strategy for the synthesis of new fully decorated 1,2,3-triazoles containing coumarin and sulfonyl-benzimidazole, and demonstrated their potential anticancer activity against various human cancer cell lines. Specifically, two compounds showed superior activity compared to standard drugs, and further experiments revealed their potency against the tyrosine kinase EGFR. In silico studies also provided valuable insights into the interactions and pharmacokinetic profiles of these compounds.

These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
