Novel avenues to control blood pressure: targeting the renal lymphatic system

Andreia Zago Chignalia

Summary: Hypertension is linked to immune and lymphatic system activation as well as lymphangiogenesis. Renal lymphangiogenesis has been shown to effectively reduce systemic arterial blood pressure in hypertensive mice. This article provides a review of the relationship between immune and lymphatic system activation and its impact on systemic blood pressure, summarizes the findings of Goodlett and collaborators, and discusses the implications of their findings.

Hydrogen Sulfide Modulates Endothelial-Mesenchymal Transition in Heart Failure

Zhen Li et al.

Summary: This study reveals that endothelial CSE modulates endothelial-mesenchymal transition and improves the severity of pressure-overload-induced heart failure by regulating nitric oxide-related mechanisms. Decreased CSE expression leads to increased endothelial-mesenchymal transition, impaired nitric oxide bioavailability, and worsened cardiac fibrosis, function, and exercise capacity. In contrast, overexpression of CSE in endothelial cells enhances nitric oxide production, reduces endothelial-mesenchymal transition and cardiac fibrosis, and improves cardiac and endothelial function, as well as exercise capacity. These findings highlight the potential therapeutic role of endothelium-derived hydrogen sulfide in treating heart failure with reduced ejection fraction.

Hydrogen Sulfide Attenuates Lymphedema Via the Induction of Lymphangiogenesis Through a PI3K/Akt-Dependent Mechanism

Junya Suzuki et al.

Summary: This study found that hydrogen sulfide promotes the growth of lymphatic vessels and ameliorates secondary lymphedema. The mechanism involves the activation of the Akt pathway in lymphatic endothelial cells. These findings provide a theoretical basis for the use of hydrogen sulfide donors in the treatment of refractory secondary lymphedema.

Role of R-spondin 2 in arterial lymphangiogenesis and atherosclerosis

Summary: RSPO2 inhibits lymphangiogenesis via LGR4 and downstream impairment of AKT-eNOS-nitric oxide signalling. These findings may inform new therapeutic strategies to promote lymphangiogenesis and improve cholesterol efflux from atherosclerotic arteries.

Oxidized Low-Density Lipoprotein Drives Dysfunction of the Liver Lymphatic System

Matthew A. Burchill et al.

Summary: This study focuses on how normal liver functions are affected during the development of nonalcoholic steatohepatitis (NASH) and the impact of inflammatory mediators on lymphatic vasculature in the liver. The findings suggest that in NASH, there is an expansion of the lymphatic vasculature which leads to downregulation of genes essential for lymphatic identity and function. Furthermore, exposure to oxidized low density lipoprotein (oxLDL) reduces lymphatic permeability in the liver, resulting in decreased protein homeostasis and increased inflammation during chronic liver disease.
